荧光定量PCR检测人类乳头瘤病毒在尖锐湿疣中的应用

摘　　要：目的　对尖锐湿疣 (CA)患者进行乳头瘤病毒 (HPV )DNA定量测定。方法　用聚合酶链反应 (PCR )测定HPVDNA的载量及HPV分型。结果　 2 0 0例尖锐湿疣患者中 198例疣体组织HPV测定阳性 ,2例阴性。在 84例疣体组织HPV6、11阳性者中有 12例合并HPV16、18阳性。 5例疣体组织及宫颈分泌物同时阳性。定量测定结果显示 :HPV6、11患者最高 2 .0× 10 8copy/ml ,最低 3 .0× 10 4copy/ml,HPV16、18患者最高 1.75× 10 7copy/ml,最低 3 .0× 10 5copy/ml。 结论　尖锐湿疣患者应用荧光定量PCR检测HPV阳性率高 ,并可快速区分致癌型及致疣型HPV感染 。Objective To detect human papilloma viruse(HPV) by the fixed quantity of deoxynycleic acid(DNA) in condyloma accuminatum patients.Methods The fixed quantity of DNA and the type of HPV was detected by PCR.Results Among the 200 condyloma accuminatum patients,198 cases were HPV16,11positive and the remaining 2 were negative in papilloma tissue. Of 84 cases,which were HPV6,11 positive in papilloma tissue,12 cases combined with HPV16,18 positive.And 5 cases had been found positive in both papilloma tissue and cervical secretion.This detection of fixed quantity showed:patients of HPV6,11 had 2.0 × 10 8 copy max and 3.0×10 4 copy min,and the ones of HPV16,18 had 1.75× 10 7 copy max,3.0×10 5 copy min.Conclusion The positive rate of HPV detected with the fixed quantity of PCR in condyloma accuminatum patients is higher,and can quickly classify whether the infected HPV is cancer type or condyloma one.In this regarding we have the possibility to forcast the recurrence and canceration of condyloma accuminatum.
