摘  要:目的 :研究桦木酸及其衍生物 2 3 羟基桦木酸等对人黑色素瘤细胞A3 75、小鼠黑色素瘤细胞B16生长抑制及凋亡作用。方法 :应用四甲基偶氮唑盐比色 (MTT)法、光镜与电镜、琼脂糖凝胶电泳和流式细胞仪等对 2 3 羟基桦木酸诱导的黑色素瘤A3 75、B16细胞进行检测和观察。结果 :桦木酸及其衍生物 2 3 羟基桦木酸等在质量浓度为 8~ 80mg·L-1时对黑色素瘤细胞有显著的生长抑制作用 ,并有一定的剂量依赖关系 ,同时出现凋亡峰及梯形电泳。结论 :2 3 羟基桦木酸能强烈诱导黑色素瘤细胞凋亡 ,从而达到抑瘤的治疗效果。objective To study the effect of apoptosis and growth inhibition induced by betulinic acid and derivatives on human melanoma A 375 cells and mouse melanoma B 16 cells.Methods MTT assay,cell morphology,agarose gel electrophoresis and flow cytometry were used to examine apoptosis and growth inhibition induced by betulinic acid and derivatives in A 375 cells and B 16 cells.Results The betulinic acid and derivatives markedly inhibited the growth of A 375 cells and B 16 cells in dose dependent manner within a given dose range (8~80?mg·L -1 ),moreover,when cells were treated with the betulinic acid and derivatives,DNA ladder with agarose gel electrophoresis and distinct apoptosis peak displayed DNA histograms by flow cytometric analysis were observed.Conclusion The betulinic acid and derivatives can strongly induce the apoptosis of B 16 cells and inhibit the growth of A 375 cells and B 16 cells in vitro .
