摘 要:目的 制备小肠粘膜下层 ,用作肌腱替代物 ,观察其替代肌腱的过程和结果 ,检验体内组织工程理论的可行性。方法 用猪小肠粘膜下层制成SIS ,用制成的SIS替代兔跟腱。观察期为 1、4、8、12、16周 ,各观察期处死 4只动物 ,进行组织学检查和力学测试 ,并对其结果进行分析和统计检验。结果 SIS替代肌腱的过程是代谢过程 ,经过 16周 ,SIS为宿主肌腱完全替代 ,其组织结构和力学性能与正常肌腱极其接近。结论 体内组织工程理论至少在肌腱再生方面是可行的。Objective To study the effect of substitution of porcine small intestinal submucosa (SIS) for rabbit Achiles tendon. Methods Porcine SIS was taken out and processed. Part of Achiles tendons of 20 rabbits′ right legs were removed and substituted by porcine SIS and the Achiles tendon of the left legs were used as controls. One, four, eight, twelve, and sixteen weeks after the operation 4 rabbits were killed and their right Achiles tendons were taken out to be examined histologically and their maximum load was tested. Results One week after the operation, the porcine SIS was already fused with the remaining part of rabbit Achiles tendon. Sixteen weeks after all the Achiles tendons looked like normal one. The maximum load of experimental Achiles tendon was 48 N±9 N one week after the operation, and increased gradually. In the 16 th week after the operation, the maximum load was 178N±6N for the experimental Achiles tendon and 174 N±10 N for the control tendon. The differences of maximun load between different weeks after operation, except that between one week and 4 weeks after, were statistically significant (P<0.01). Conclusion Substitution of porcine SIS for injured Achiles tendon is effective, thus proving the feasibility of in vivo tissue engineering technology.
