摘  要:目的 研究二乙酰二脱水卫矛醇 (1,2 :5 ,6 -dianhydro - 3,4-diacetylgalactitol,DADAG)的抗脑白血病作用及机制。方法 用小鼠脑内移植瘤模型、MTT法和DNA掺入法 ,观察DADAG对小鼠脑内移植瘤和体外白血病L12 10细胞的作用 ,并探讨作用机制。结果 DADAG对DBA/ 2小鼠脑内移植白血病L12 10细胞有明显的抑制作用 ;对体外白血病L12 10细胞同样有很强的抗增殖作用 ,其IC50 值为 2 4.6mg·L-1。DADAG不可逆地抑制L12 10细胞内DNA的生物合成。Objective To study the inhibitory effects of diacetyldianhydrogalactitol (DADAG) on brain leukemia and its mechanism. Method To observe the antitumor effect of DADAG in vivo using intracerebrally inoculated tumor cells in mice model. The effect of DADAG on the proliferation of mouse L1210 leukemic cells were observed with MTT assay and incorporation of TdR into DNA of the cells. Results DADAG exhibited significant antitumor effect on intracerebrally inoculated L1210 leukemia in DBA/2 mice. DADAG showed potent antiproliferative effect on L1210 cells in vitro and the IC 50 of DADAG was 24.6?mg·L -1 in MTT experiment. It also irreversiblely inhibited the incorporation of TdR into DNA of L1210 cells. Conclusion The antitumor effect of DADAG on brain leukemia is related to its remarkable inhibitory effect on proliferation and DNA synthesis of leukemic cells.
