多囊平煎剂组方对大鼠高睾酮血症影响的正交设计

摘　　要：目的　观察多囊平煎剂中各味药物对模型大鼠高睾酮血症的血清睾酮的影响 ,探讨多囊平煎剂对高睾酮血症的最佳药味组合 ,说明该方中药物作用的主次与配伍规律。方法　采用正交设计进行拆方研究 ,确定因子与水平数 ,取给药与不给药两水平 ,给药剂量为原方剂量 ,按正交表L16(2 15)的设计 ,将大鼠分为 16个组 ,每组重复 3次 ,分别给予不同的方剂组合并观察大鼠用药后的血清睾酮值。结果　多囊平煎剂中单味药以赤芍降低睾酮作用最显著 (P <0 0 1) ,依次为瓦楞子、甘草、单味药白芍降低血清睾酮作用不显著 ,但交互作用分析以白芍与甘草合用降低血清睾酮作用最显著 (P <0 0 1) ,其次为赤芍与甘草合用 (P <0 0 5 )。泽兰降低血清睾酮作用不显著 ,与其他药交互作用降低血清睾酮作用不显著。结论　多囊平煎剂中降低血清睾酮作用最显著 ,即最优药味组合为白芍、甘草、赤芍、瓦楞子。Objective To find out the optimal composition of duonangping decoction (DNP);at present it consists of five medicinal herbs to treat hyperandrogenemia with a good effect.Method Orthogonal design and a “dismemberment method”were taken in this study,according to orthogonal layout L 16(2 15),rats were divided into 16 groups,and were given doses of the dismembered DNP prescription (ie,different herb matching)for three times to observe the serum levels of testosterone in rats.Results Among the DNP′s ingredients the herb Radix paeoniae rubra lowered the testosterone level (T level) most powerfully (P<0.01),the less powerful were Concha arcae and Radix glycyrrhizae;single Radix paeoniae alba did not lower the T level markedly.But interactional analysis revealed that a match of Radix paeoniae alba and Radix glycyrrhizae exerted a strongest action to lower T level(P<0.01),the stronger were Radix paeoniae rubra and Radix glycyrrhizae in a match(P<0.05).Herba lycopi had little action on T level,its match with other herbs also had no noticeable effect on T level.Conclusion The optimal prescription of duonangping decoction is the composition of Radix paeoniae alba.Radix glycyrrhizae,Radix paeoniae rubra,and Concha arcae,which showed the best action to lower T level;Herba lycopi showed little action as such.
