摘  要:①目的 探讨乙醇对胚胎的致畸作用 ,并建立一个较理想的乙醇致畸动物模型。②方法 以金黄地鼠为实验动物 ,以体积分数为 0 .2 5的乙醇生理盐水溶液为致畸原 ,以多个不同剂量 (2 .0 ,4 .0 ,5 .8和 7.0 g/kg体质量 )、不同给药时间 (孕 6~ 8d)的组合进行致畸效果观察。③结果 给药时间为孕第 7.5天时 ,随给药剂量升高 ,吸收胎 +死胎率及畸胎率明显升高 ,其中 5 .8及 7.0 g/kg体质量组吸收胎 +死胎率分别为 34.15 % ,77.0 8% ,畸胎率分别为 75 .6 8% ,81.81% ;给药剂量固定为 5 .8g/kg体质量时 ,各时间组畸胎率与对照组相比差异均有统计学意义 (χ2 =11.2 2~ 78.0 2 ,P <0 .0 0 1) ,但主要畸形种类不同 ,其中孕第 7.0~ 7.5天出现严重的神经管畸形和眼畸形。④结论 孕第 7.5天腹腔注射 5 .8g/kg体质量乙醇溶液 ,畸胎率较高 ,而吸收胎 +死胎率相对较低 ,畸形严重 ,是一个较理想的乙醇致畸模型。Objective\ To study the teratogenic effects of ethanol on embryos and to establish a good animal model of ethanol induced malformation. \ Methods\ Pregnant golden hamsters were given a different single dose of 25%( V/V ) ethanol saline solution (5.8 or 2.0, 4.0, 7.0 g/kg bodyweight) on different gestation days (GD 6.0, GD 7.0, GD 7.5 or GD 8.0). On GD 13.0 , the dams were killed and the teratogenic results were examined. \ Results\ When the dams were ethanol exposed on GD 7.5 , the percentage of resorptions+deaths and malformed fetuses were increased in a dose related manner. When the dose was 5.8 g and 7.0 g/kg bodyweight, the resorption and death rates were 34.15%, 75.08%, the malformation rates were 75.68% and 81.81% , respectively. When the dose of 5.8 g/kg bodyweight was given for various groups ,the malformation rate was with statistic significance as compared with the control groups( χ 2=11.22-78.02,P <0.001). But in different days, the major malformations were different. On GD 7.0, 7.5, there were severe neural tube and eye malformations. \ Conclusion\ Given 5.8 g/kg bodyweight ethanol solution abdominally on the GD 7.5 of the golden hamsters can cause higher teratogenic rate, relatively lower resorption rate and more severe malformations. So it is a good animal model of ethanol induced malformation.
