摘 要:光敏核不育水稻可育花粉单核中位期外壁覆盖层和外壁内层界限明显 ,ATPase定位显示覆盖层外侧和花粉核有活跃的ATPase活性。单核边位期ATPase定位于覆盖层外侧、柱状层、内壁、线粒体以及核仁上 ;二核期ATPase定位于覆盖层外侧、柱状层、内壁和核。不育花粉单核中位期外壁发育不良 ,花粉核形状不规则 ,有明显核仁 ,核仁ATPase活性较核其余部分强 ;单核边位期不育花粉仍不能形成良好的外壁 ,ATPase主要定位于核、内壁和线粒体 ,覆盖层外侧和不规则的柱状层也具有ATPase活性。二核期不育花粉ATPase定位于内壁和变形的柱状层。可育花药单核中位期绒毡层ATPase定位于核和线粒体 ,同期不育花药绒毡层ATPase定位于核和核仁上。部分不育花药药隔维管束也表现异常。基于以上结果 ,认为不育花药绒毡层、单核中位期花粉核异常可能与败育有较密切的关系。ATPase activity of in ‘Nongkeng58s’ (rice, Oryza sativa L.) anthers was detected with Pb 3 (PO 4) 2 precipitation technique during different developmental stages of pollen. At small-vacuole pollen stage, fertile pollen (Nongkeng58s-SD, short daylight) showed well developed tectum, columellae and endexine, and ATPase was mainly localized in nucleus and on the surface of tectum. While at big-vacuole pollen stage, fertile pollen showed ATPase activity in nucleolus, mitochondria, intine, columellae and on the surface of tectum. At bi-nucleate pollen stage, ATPase was localized in nucleus, endintine, columellae and on the surface of tectum. In sterile pollen (Nongkeng58s-LD, long daylight), exine did not develop well at small-vacuole stage, and ATPase activity in nucleolus was more active than that of the rest of nucleus. The exine of sterile pollen was not perfectly differentiated even at big-vacuole pollen stage, and ATPase was localized in nucleus, mitochondria, intine, malformed columellae and on the surface of the tectum. At bi-nucleate pollen stage, sterile pollen showed ATPase activity in intine and distorted columellae. The tapetum of fertile anther showed ATPase activity in mitochondria and nucleus, while in sterile anther, tapetum exhibited ATPase activity only in nucleus. In addition, the connective structure of sterile anther was different from its counterpart in fertile anther. Based on the above results, we supposed that malfunction of tapetum and nucleus in sterile pollen might be closely related to male sterility.
