摘 要:目的 观察血性脑脊液(BCSF)对内皮细胞(EC)分泌内皮素(ET)和一氧化氮(NO)的影响及川芎嗪(LZ)、尼莫地平(NM)的干预作用。方法建立脐静脉EC体外培养的方法,测定ET、NO的代谢产物NO_X- 含量,并观察BCSF和NM及LZ的作用。结果LZ和NM可抑制正常体外培养的EC分泌ET,对NO无影响;不同时间BCSF均使ET分泌增加,NO分泌减少;LZ既能减少BCSF引起的ET升高,又可抑制NO的下降,而NM仅能抑制ET的增高,对NO无明显影响。结论蛛网膜下腔出血(SAH)后,通过破坏ET/NO的平衡,导致脑血管痉挛(CVS)。LZ和NM可通过抑制这一作用而防治CVS。Objective To study the effect of BCSF on the ET and NO from human EC and pharmacologial intervention of LZ and NM. Method The EC cultures from human unbilical vein were established. Concentration of ET and NO were determined using radioimmunoassy and HNO<sub>3</sub> reductase respectively and effects of BCSF, LZ and NM were conducted. Result ET secretion farm nomal EC could be inhibited by LZ and NM wheras NO yield was not affected. ET elevaton induced by BCSF could be inhibited by LZ as well as NM while NO reduction could only be inhibited by LZ. Conclusion LZ and NM could be used in prevention and treatment CVS following subarachnoid hemorrage(SAH) by inhibiting ET elevation and NO reduction.
