摘  要:目的:探讨雷帕霉素对K562细胞survivin、caspase-3在mRNA水平的表达和对K562细胞超微结构的影响。方法:用不同浓度雷帕霉素处理慢性髓系白血病K562细胞,应用CCK8的方法检测雷帕霉素对K562细胞的增殖抑制率,应用电子显微镜观察K562细胞形态学的改变,应用RT-PCR的方法检测不同浓度雷帕霉素作用K562细胞后survivin,caspase-3在mRNA水平的表达情况。结果:雷帕霉素对K562细胞的增殖有明显的抑制作用;电子显微镜显示,随着雷帕霉素浓度的增高K562细胞的凋亡程度加重;随着雷帕霉素浓度的增高caspase-3在mRNA表达水平上升,而survivin在mRNA水平表达下降。结论:雷帕霉素通过上调caspase-3的水平,可以降低survivin水平而促进K562细胞的凋亡。Objective:To investigate the effect of rapamycin on the expression of survivin and caspase-3 at mRNA level in K562 cells and the influence of rapamycin on K562 cell ultrastructure.Methods:The effects of rapamycin at various concentration on K562 cell proliferation were analyzed by CCK8;the morphological characteristics of K562 cells was observed by transmission electron microscopy;the expression of survivin and caspase-3 at mRNA level in K562 cells treated with rapamycin was detected by RT-PCR.Results:The proliferation of K562 cells was significantly inhibited by rapamycin.The apoptosis level of K562 cells increased with increase of rapamycin concentration,the expression of survivin at mRNA level decreased with increase of rapamycin concentration(P <0.05).The expression of caspase-3 at mRNA level increased with increase of rapamycin concentration.Conclusion:Rapamycin can promote K562 cell apoptosis through up-regulating caspase-3 level and reduceing survivin level.
