摘 要:本文报告给犬经气道和静脉应用尼可刹米。结果显示经气道和静脉应用同等剂量的尼可刹米(8.3mg/kg),其血药浓度在1.5分钟后差异不显著(P>0.05)。经气道应用高剂量尼可刹米时(25mg/kg),其血药浓度随之升高。经气道和静脉用药后,呼吸频率、潮气量均增加。经气道用药后半吸收期为0.48分钟,3小时内生物利用度为86%。以上结果提示,经气道应用尼可刹米能经肺部迅速吸收并能达到与静脉应用等剂量药物时的相同药理效应。Studies have been conducted to determine whether nikethamide may be effectively and safely absorbed through the endotracheal route in the canine model. As the results, one and half(1.5)minutes after administration of nikethamide the difference of drug concentration following the intravenous and endotracheal route (8.3mg/kg) was not significant. However, the blood concentration of nikethamide is increased when the endotracheal dose was inereasesed, 50% being absorbed 0.48 minutes after endotracheal administration. Bioavailability is 84.69%. The frequency of breath and tidal volume were increased after the administration of nikethamide. These results suggest that endotracheal application of nikethamide can produce the similar effects as through the intravenous route.
