摘  要:构建植物表达载体并利用该载体与含有选择压力基因的载体对小麦幼胚愈伤组织进行共转化。比较分析了高渗透压处理阶段使用0.4 mol/L的蔗糖(合12%)代替甘露醇及恢复培养阶段使用过渡浓度的蔗糖(6%,介于高渗透压培养基中蔗糖的浓度12%和分化培养基中蔗糖的浓度3%之间)对分化率和转化效率的影响。结果表明,在采用基因枪轰击法叶小麦幼胚愈伤组织进行转化的体系中,以蔗糖代替甘露醇进行高渗透压处理,进而使用含有较高浓度蔗糖的培养基进行恢复培养的方法简便有效,可以促进分化能力提高,转化效率可达2.1%。The wheat immature embryos were bombarded with plasmids containing the plant expression vector, which was constructed in this experiment. The authors evaluated the impact on regeneration efficiency and transformation efficiency by using 0. 4 mol/L sucrose (means 12 percent) instead of mannitol during the osmotic pretreatment as well as using 6 percent sucrose during the recovery culture. The results showed that the method applying sucrose in pretreatment medium and using recovery medium with intermediate high contents of sucrose was both efficient and convenient. The regeneration capacity had been improved and the transformation efficiency reached 2. 1 percent.
