摘 要:将 2~ 3个外源目的基因与筛选标记基因hpt连在同一个载体上 ,用基因枪法对 1个粳稻品种和 4个籼稻品种进行了转化。粳稻中花 8号转化频率最高为 5 6 .4% ,籼稻七丝软占、特青、奇妙香、九六零转化频率分别为 6 .4% ,2 .5 % ,2 .8%和 1.7%。分子杂交实验证明 ,通过两次潮霉素抗性筛选的T0 代再生植株 10 0 %含有hpt基因 ,其中 70 %以上含有外源目的基因的完整表达结构。遗传分析表明 ,大部分T1代植株中潮霉素抗敏比符合孟德尔We constructed two or three target genes and the marker gene hpt into one vector(Fig.1), and transformed them into one indica rice variety and four japonica rice varieties(PlateⅠ a,b,c,d,e,f). Among them, the Zhonghua No.8 had the highest transformation rate 56.4%, and the transformation rate of Qisiruanzhan, Teqing, Qimiaoxiang and Jiuliuling is 6.4%, 2.5%, 2.8% and 1.7%. Molecular analysis proved that 100% of the regenerated plants contained hpt transgenes after hygromycin selection for two times, and more than 70% of them contained intact expression construct of foreign target gene(Figs.2,3). Genetic analysis showed that the hyg r/ hyg s ratio in most of T 1 plants is near 3 to 1(PlateⅠ g,h), corresponding to Mendelian ratio.
