摘  要:目的:研究原钒酸钠对2型糖尿病大鼠丙酮酸激酶活性的影响。方法:大鼠灌胃给予脂肪乳10d后,应用四氧嘧啶建立2型糖尿病模型。70只大鼠分为正常组、高脂组、糖尿病组、苯乙双胍组和原钒酸钠大、中、小剂量组(9、3、1mg.kg-1)。连续灌胃7d后,测定各组空腹血糖值,用比色法测定肝组织和血清中丙酮酸激酶活性。结果:原钒酸钠降低2型糖尿病大鼠血糖值;与模型组相比,中、大剂量原钒酸钠升高2型糖尿病大鼠丙酮酸激酶活性(P<0.01)中剂量原钒酸钠使血清中酶活性完全恢复,大剂量原钒酸钠使肝组织和血清中酶活性完全恢复(与正常组相比P>0.05)。结论:原钒酸钠增加2型糖尿病大鼠肝组织和血清中丙酮酸激酶活性,这可能是其发挥降血糖作用的一个环节。Objective:To determine the effects of sodium orthovanadate on pyruvate kinase of type II diabetes rats.Methods:A type II diabetic rat model was established following daily oral consumption of high concentration of the fat diet for ten days and then injection(ip)with alloxan.The fasting blood sugar,the activity of pyruvate kinase in liver tissue and serum,serum nitric oxide(NO)and insulin levels were evaluated in seven groups of rats(n= 70)receiving either normal diet(healthy control),highly fatted diet,phenformin,or sodium orthovanadate at doses of either 0(normal diet control),1,3 or 9 mg·kg-1,ingested as part of their daily diet for 7 days.Results:The fasting blood sugar level was declined at all sodium orthovanadate dose-treated groups.The activity of pyruvate kinase was elevated at the sodium orthovanadate doses of 3 and 9 mg·kg-1 groups compared with the one in the normal diet group(P<0.01).The pyruvate kinase values were back within normal range in the serum when the rats were treated with sodium orthovanadate dose of 3 mg·kg-1,and in the liver tissue and the serum at 9 mg·kg-1 compared with the one in healthy control group(P>0.05).The NO generation was inhibited by sodium orthovanadate at the doses of 3 and 9 mg·kg-1.No significant effects on the insulin level were observed in all sodium orthovanadate dose-treated groups.Conclusion:Sodium orthovanadate might play a role in the reduction of hypeglecemia by the elevation of pyruvate kinase activity in type II diabetes rats.
