摘 要:目的 研究维生素 A的致畸性和锌对维生素 A致畸作用的影响。方法 将不同剂量的维生素A(0、2 .5、5、10、15μg/ ml)加入培养基中 ,或同时加入 5μg/ ml维生素 A和不同剂量的锌 (5、10、15μg/ m l) ,用计算机图像分析仪观测培养后肢体的发育和分化情况。结果 随着维生素 A剂量的增加 (2 .5~ 15 .0μg/ m l) ,肢体中各软骨原基的发育和分化越差、面积越小 ,并对软骨原基的外形发育产生抑制。长骨和爪骨都受到严重影响 ,对骨组织的作用比对软组织大 ,后肢畸形比前肢严重。当同时加入的锌量增加时 (5~ 15μg/ ml) ,肢体中各软骨原基的发育和分化越差、面积越小。结论 过量维生素 A具有较强的致畸性 ,而过量的锌不能改善过量维生素 A的致畸作用 。Objective To search into the teratogenicity of vitamin A and the influence of zinc on the teratogenesis induced by vitamin A. Methods The 12 day old mouse embryo limb buds were respectively cultivated in the basic culture medium with Vit.A of 0, 2.5, 5.0, 10.0, 15.0 μg/ml or in the basic culture medium with Vit.A of 5.0 μg/ml and Zn of 5.0, 10.0, 15.0 μg/ml. After 3 days' incubation, the morphogenetic differentiation of the cultured limb was examined and the area and forms of the bone anlagen were measured by image analyzer. Results Vit.A could induce micromelia and digital defect (ectrodactylia and syndactylia) in limbs. As the dose of Vit.A increased, the lesions also increased and a dose dependent decrease was observed in the area of bone anlagen both in fore and hind limbs. The effect was more severe in hind limbs, paws and long bones. Vit.A had a greater effect on chondrogenic tissue than on soft tissue. When various levels of Zn were added to the culture medium along with Vit. A (5.0 μg/ml), cultured limbs were affected more severely than those caused by Vit.A alone. Conclusion Large doses of Zn can not mitigate the detrimental effect of excessive intake of Vit.A; on the contrary, over doses of Zn will exacerbate the teratogenic effect of over dosed Vit.A.
