摘 要:目的 探讨一氧化氮 (NO)、铁调节蛋白 (IRP)在慢性病贫血 (ACD)大鼠肝组织铁代谢紊乱中的调节作用。方法 在传统佐剂性关节炎大鼠模型基础上 ,通过追加注射福氏佐剂两次 ,增强其免疫反应 ,从而诱发建立贫血大鼠模型。对贫血模型组和正常对照组大鼠肝脏铁含量、铁蛋白、NO、IRP- 2 m RNA水平及 IRP结合活性进行检测。结果 佐剂性关节炎大鼠经追加福氏佐剂 ,肝铁、铁蛋白水平升高 (P<0 .0 5〉,与 ACD铁代谢改变特征一致 ;ACD大鼠肝组织 IRP结合活性较正常对照组升高 ,IRP- 2 m RNA表达下调 ,NO水平与 IRP结合活性呈正相关。结论 NO、IRP在Objective To investigate the regulatory effect of Nitrogen Oxide (NO) and Iron Regulatory Protein (IRP) on the iron metabolism disorder in the livers of rats with Anemia of Chronic Disease (ACD). Methods Anemia was induced by injecting freund adjuvant twice to the traditional model rats of adjuvant arthritis to enhance their immune responses. We compared the levels of iron, Ferritin, NO, and the IRP binding activity and the expression level of IRP 2 mRNA in the livers of rats between the experimental group and the normal group. Results The levels of iron and Ferritin in the livers of adjuvant arthritis rats elevated by injecting the Freund adjuvant, which was consistent with the characteristics of iron metabolism disorder in ACD. The IRP binding activities in the livers of ACD rats were greater than those of the control group, while the expression of IRP 2 mRNA decreased. NO was positively correlated to the IRP binding activities. Conclusion NO and IRP play an important role in the disorder of iron metabolism in the livers of ACD rats.
