摘 要:目的 :探讨人血管平滑肌细胞的低密度脂蛋白受体 (LDL -R)的表达与神经肽Y(NPY)之间的关系 ,以阐明NPY在动脉粥样硬化的发生发展中的重要地位。方法 :将第 3~ 4代融合生长的hVSMC分成10 -6mol/LNPY组和对照组 ,分别培养 6 ,12 ,2 4 ,4 8h四个时间段后 ,进行荧光免疫组化染色 ,然后在激光扫描共聚焦显微镜系统下 ,定量观察NPY对人血管平滑肌细胞的LDL -R表达的影响。结果 :各时间段中对照组hVSMC的LDL -R表达的平均荧光值分别为 2 5 2 7.4 5± 2 0 9.2 2 (6h) ,2 4 76 .6 6± 2 10 .2 8(12h) ,2 4 4 2 .6 5±189.16 (2 4h)和 2 4 2 8.39± 191.0 3(48h) ,各时间段之间均无显著性差异 (P >0 .0 5 ) ;而各时间段中NPY组hVSMC的LDL -R表达的平均荧光值分别为 2 2 80 .6 8± 174 .5 7(6h) ,1878.81± 15 4 .18(12h) ,184 0 .78±12 3.2 9(2 4h)和 172 1.6 8± 14 3.4 5 (48h) ,与相应时间段的对照组相比分别下降 9.76 % ,2 4 .14 % ,2 4 .6 4 %和2 9.10 % (P <0 .0 1) ,且各时间段之间有显著性差异 (P <0 .0 5 ) ,呈现一定的时间依赖效应。结论 :人血管平滑肌细胞的LDL -R表达受NPY下调作用的影响 ,可导致LDL -R介导途径障碍 ,从而使LDL在血管壁中沉积 ,这可能是NPY参与动脉粥样硬化发生发展的重要机制之一?Objective:To explore the relationship between the expression of low density lipoprotein receptor (LDL-R) in human vascular smooth muscle cells (hVSMC) and neuropeptide Y (NPY) so as to elucidate the significant role of NPY in the occurrence and progression of Atherosclerosis.Methods:The 3rd~4th generations of syncretic hVSMCs were divided into the control groups and 10~ -6mol/L NPY groups and were cultured for 6h, 12h, 24h and 48h respectively, then, were stained by the immunofluorescent histochemical staining and quantitatively assayed the effect of neuropeptide Y on the expression of LDL-R in human vascular smooth muscle cells under laser scanning confocal microscope.Results: In different culture time, the average fluorescent values of the LDL-R expression in hVSMC of control groups were 2527.45±209.22(6h), 2476.66 ± 210.28(12h), 2442.65±189.16(24h) and 2428.39 ±191.03(48h) respectively, and there were not significant differences among them (P> 0.05). While the average fluorescent values of the LDL-R expression in hVSMC of NPY groups were 2280.68±174.57(6h), 1878.81±154.18(12h), 1840.78±123.29(24h) and 1721.68±143.45(48h), decreasing by 9.76%, 24.14%, 24.64% and 29.10% respectively compared with corresponding control group (P>0.01). There were significant differences among the different action time NPY groups in a dose-dependent manner (P<0.05).Conclusion:The expression of LDL-R in hVSMCs was downregulated by NPY. It probably resulted in blocking the LDL-R-mediated pathway, consequently LDL could accumulated in the vascular wall. This might be the important mechanism that NPY participate in the occurrence and progression of atherosclerosis.
