摘 要:目的 探讨饮用水有机污染物的潜在生殖危害。方法 用GDX 10 2树脂吸附饮用水中有机污染物 ;昆明种雄性小鼠分为对照组及 12 .5、2 5、5 0L/kg试验组 ,腹腔注射染毒 ,分别于染毒后 10、3 5d处死动物取材 ,应用光镜和电镜技术对睾丸、附睾、精囊、前列腺进行组织病理学研究。结果 染毒后 10d ,仅发现 5 0L/kg组间质细胞增生 ,坏死生精细胞较多 ;染毒后 3 5d ,5 0L/kg组间质细胞呈大片聚集状增生 ,曲细精管萎缩 ,生精细胞空泡化。电镜观察所见 ,染毒后 10d ,5 0L /kg组Leydig细胞线粒体肿胀、嵴紊乱 ;染毒后 3 5d ,Leydig细胞、生精细胞损伤随剂量增加而加重 ,损伤以线粒体的肿胀、空泡化甚至溶解为主。附睾、精囊、前列腺均未发现异常形态学变化。结论 饮用水有机污染物存在生殖危害 ,睾丸是其作用靶器官 ,靶细胞为Leydig细胞和生殖细胞 。Objective To explore the potential reproductive toxicity of organic pollutants in drinking water. Methods Organic pollutants were extracted with GDX 102 resin from drinking water and then injected intraperitoneal to male mice in different dosage of 12.5, 25 and 50 L/kg for 5 d. On the 10th and 35th day after treatment, the mice were killed and the testis, epididymis, seminal vesicles and prostate gland were removed and fixed for pathohistological examination. Results On the 10th day after treatment, hyperplasia of Leydig cells and increase of necrotic spermatogenic cells were found only in the mice treated at the dosage of 50 L/kg; On the 35th day, the Leydig cells in these mice demonstrated massive accumulating hyperplasia, the seminiferous tubules shrunk and spermatogenic cells were vacuolated. Under electron microscope, the mitochondria of Leydig cells were swellen and with disordered ridge in 50 L/kg mice 10 d after treatment, and on the 35 th day, the damage of Leydig cells and spermatogenic cells were more serious in swelling, vacuolation and lysis. Conclusion The results suggest that the organic pollutants in drinking water is harmful to reproductive system, with the testis as target organ, Leydig and spermatogenic cells as target cells, and their mitochondria as target site.
