摘  要:目的 研究 IL - 6对 IFN-γ、IL - 1诱导人脑膜成纤维细胞 (Human meninx fibroblasts,HMF)产生吲哚胺 2 ,3-双加氧酶 (indoleamine 2 ,3- dioxygenase,IDO)抗弓形虫作用的影响。 方法 采用3H-尿嘧啶 (3H- U)特异标记的弓形虫在HMF内的增殖实验 ,以及通过反相高效液相色谱仪检测 IDO活性。 结果  IL- 6能诱导 HMF产生 IDO降解胞内必需氨基酸色氨酸 ,由于弓形虫发生“色氨酸饥饿”而被抑制生长 ;加入抗 IL- 6抗体可逆转 IFN- γ、IL- 1对弓形虫的抑制作用。结论  IL - 6可诱导 HMF产生 IDO抑制弓形虫生长 ;IL - 1单独或协同 IFN-γ抗弓形虫增殖都是通过 IL - 6诱导 HMF产生 IDO而介导的。Objective In vitro cultured human meninx fibroblasts(HMF) were treated with IL 6, IFN γ and IL 1, the effect of IL 6 on toxoplasmastatic activity of indoleamine 2,3 dioxygenase(IDO) induced by IFN γ and/or IL 1 was studied. Methods The ability to restrict growth of Toxoplasma gondii was evaluated by 3H U incorporation assay. IDO activity was determined with high performance liquid chromatography(HPLC). Results IDO was induced by IL 6 in HMF, which could degrade the intracellular tryptophan. T. gondii growth was then restricted because of hypotryptophan during the process of protein synthesis. Addition of antibody against IL 6 to the culture medium blocked the antiparasitic effect of IFN γ and/or IL 1. Conclusion IL 6 could induce IDO to restrict T. gondii growth in HMF. The antiparasitic effect of IFN γ and/or IL 1 was mediated by IL 6 inducing IDO in HMF.
