摘  要:为了探讨小麦(Triticum aestivum)杂种优势形成的分子机理,选用杂种优势不同的3个杂交组合,以授粉后2~12 d的杂交和自交的种子为材料,采用mRNA差异显示技术分析了MADS-box和Ser/Thr蛋白激酶两个基因家族在不同优势组合的杂交和自交种子发育前期的基因表达差异.结果表明,所有6个时期杂交和自交种子之间存在明显的基因表达差异,表现为量和质的两类差异.进一步分析发现,强优势组合差异表达比例较弱优势组合高.3个杂交组合农大3159×京冬6号、农大3159×原冬8790和农大3159×农大3214与农大3159自交相比,MADS-box家族基因的差异表达比例随发育时期呈一定的递增趋势,但在授粉后4和12 d时,差异表达的比例明显减少;Ser/Thr蛋白激酶家族基因差异表达比例在授粉后2、4和6 d时随发育时期呈递增趋势,6 d时差异表达达到最大,后逐渐减小.In order to understand the molecular basis of heterosis, modified differential display of mRNA was used to characterize the difference in gene expression between cross-fertilized and self-fertilized kernels at 2, 4, 6, 8, 10 and 12 d after pollination (DAP) by using 3 wheat (Triticum aestivum) hybrids with different levels of heterosis. The results indicated that the patterns of gene expression altered obviously in cross-fertilized kernels as compared to self-fertilized kernels. Both quantitative and qualitative differences were observed. Major difference of gene expression occurred in the qualitative level. The further analysis showed that higher percentage of differentially expressed cDNA was observed in highly heterotic hybrid. As far as MADS-box concerned, with the development of seed, there were gradually higher percentage of differentially expressed cDNA in Nongda3159 × Jingdong6, Nongda3159 × Yuandong8790 and Nongda3159 × Nongda3214 as compared to self-fertilized Nongda3159, but decreased at 4 DAP, and the cross-fertilization effect was maximum at 10 DAP. As for Ser/Thr gene family,there were gradually higher percentage of differentially expressed gene at the 2 DAP, 4 DAP and 6 DAP, then decreased, and the cross-fertilization effect was maximum at 6 DAP.
