摘  要:目的 研究不同碘水平饮食对实验性自身免疫性甲状腺炎 (EAT)大鼠甲状腺的Th1型细胞因子IFN -γ和Th2型细胞因子IL - 10表达的影响 ,进一步探讨碘在自身免疫性甲状腺疾病 (AITD)发病中的作用机理。方法 雌性Lewis大鼠用低碘饲料和含不同浓度的KI水喂养 ,即分为低碘 (LI)、适碘 (NI)和高碘 (HI)组。EAT诱发组用Wis tar大鼠Tg免疫 ,诱发EAT的形成 ,观察大鼠甲状腺病理改变、血清中Tg自身抗体水平和甲状腺IFN -γ和IL -10mRNA的表达水平。结果  (1)EAT诱发的HI组大鼠甲状腺炎细胞浸润明显 ,接近重度炎症 ,NI组大鼠甲状腺为近炎症恢复期 ,而LI组呈轻度炎症。 (2 )EAT诱发组的HI、NI和LI大鼠血清中抗Tg自身抗体平均水平明显高于非EAT诱发对照组 ,P <0 0 1。 (3)EAT诱发组中的HI组大鼠甲状腺IFN -γ和IL - 10mRNA的表达 ,均明显高于非EAT诱发对照组 ,P <0 0 5。NI组大鼠甲状腺IL - 10mRNA和LI组甲状腺IFN -γmRNA的表达均高于非EAT诱发对照组 ,P <0 0 5。结论 EAT诱发组中的HI组大鼠甲状腺有较高水平的IFN -γ和IL - 10mRNA的表达 ,出现明显炎症和抗炎症反应 ;NI组大鼠仅Th2型细胞因子IL - 10较高水平表达 ,甲状腺炎症处于恢复期 ;LI组大鼠有较高水平IFN -γ的表达 ,甲状腺炎症反应较轻 。Objective In order to further understand the role of iodine in the invasion of autoimmune thyroid disease(AITD),effects exerted by different dietary iodine on the expression of Th1 type cytokine(INF-γ) and Th2 type cytokine(IL-10) in thyroid were investigated in experimental autoimmune thyroiditis(EAT) rats.Methods Female Lewis rats were fed by low-iodine diet and drunk deionized water containing different concentration of KI respectively.Correspondingly,rats were divided into LI group,NI group and HI group.Induced EAT rats were passively immunized with Wistar rats' Tg.The thyroid tissue pathological changes were studied.Serum concentration of anti-Tg autoantibodies were measured and the expression of INF-γ and IL-10 genes in thyroid was analyzed.Results (1)EAT-induced rats from HI group had a more clearly lymphocytic infiltrates indicating a more severe inflammation in thyroid glands while those from NI group appeared a mild infalmmation moreover in restoration stage and rats from LI group just showed a light lymphocytic infiltrating within follicles.(2)The titer in EAT-induced rats was much higher than that of not induced EAT-rats, P <0 01.(3)The expression of INF-γ and IL-10 in EAT-induced rats from HI were both higher than its control group, P <0 05.The expresion of INF-γ in EAT-induced,rats from NI and the expression of IL-10 in EAT-induced rats from LI were significantly increased compared with respective control group.Conclusion A much higher expression of INF-γ and IL-10 appeared in thyroid glands of EAT rats from HI group which manifested an inflammatory reaction and anti-inflammation reaction.Only high expression of Th2 type cytokine IL-10 appeared in EAT rats from NI group with an inflammation in restoration stage and the expression of INF-γ in EAT rats from LI group revealed that the infamlmation was very light and the process was slow.
