左归丸对卵巢切除所致骨质疏松大鼠IL—1和IL—6活性的影响

摘　　要：目的:探讨左归丸防治骨质疏松症的机理。方法:以卵巢切除所致的骨质疏松大鼠为模型,采用骨组织形态计量学方法测定胫骨骨小梁体积百分比(TBV%)、骨小梁吸收表面百分比(TRS%)和骨小梁形成表面百分比(TFS%),采用放射免疫分析法测定外周血清中雌二醇(E_2)含量,分别采用小鼠胸腺细胞检测法和依赖株细胞增殖反应法测定白细胞介素-1(IL-1)和白细胞介素-6(IL-6)的活性。结果:左归丸能使胫骨TBV%显著增高,使TRS%和TFS%显著降低。大鼠切除卵巢后,E_2含量大幅度降低,而IL-1和IL—6活性显著增高。左归丸对E_2含量无显著性影响,但对IL—1和IL—6活性有明显的抑制作用。结论:左归丸能够抑制IL-1和IL-6的活性是其防治骨质疏松症的机理之一。Purpose: To explore the mechanism of Zuo Gui Pill for treatment of osteoporosis. Methods: In the vat of osteoporosis induced by ovariectomy. tibial bone trabecula volume percent (TBV%), tibial bone trabecula absorption percent (TRS%) and tibial bone trabecula forming surface percent (TFS%) were determined with bone tissue morphometry, and peripheral serum estradiol (E2) content was determined by radioimmunoassay. and interleukin-1 (IL - 1) and IL - 6 activities were determined with mouse thymocyte detection and dependent strain cell proliferation reaction method. Results: Zuo Gui Pill could increase significantly TBV%. decrease TRS% and TFS%. After ovariectomy. Ez decrease at a large amplitude greatly and IL-1 and IL-6 increased significantly, and Zuo Gui Pill did not have significant effect on E2 content, but had a significant inhibition on IL - 1 and IL - 6 activities. Conclusion: Zuo Gui Pill can inhibit II, - 1 and IL-6 activities which is one of the mechanism for prevention and treatment of osteoporosis.
