摘　　要：目的探讨IGF-Ⅰ与其受体两亚基(α,β)在正常皮肤和溃疡中的分布和表达特征。方法溃疡8例和正常皮肤8例用免疫组化方法确定3种蛋白定位和表达量。结果在溃疡组织中,与正常皮肤相比IGF-Ⅰ表达虽有所升高,但增加不显著(P>0.05),蛋白定位差异无显著性意义。含有IGF-ⅠRα和IGF-Ⅰβ蛋白的阳性细胞主要为部分表皮细胞、单核细胞和巨噬细胞,两种蛋白的阳性表达率分别降至正常皮肤的43.9%和50.0%(P<0.05)。结论在IGF-Ⅰ因子高浓度环境中,溃疡创面难愈性修复可能与IGF-Rα和IGF-Ⅰβ蛋白表达下降,引起因子与受体结合发生障碍相关。Objective To investigate the expression andlo cation of insulin-like growth facto r-Ⅰ(IGF-Ⅰ)and two units of its receptor IGF-ⅠRαand IGF-ⅠRβin dermal chronic ulcers and normal s kins in order to explore their intluences on ulcer formation.Methods The expression intensity and distribut ion of IGF-Ⅰ,IGF-ⅠRαand IGF-ⅠRβwere detected with patholical method and immunohistochemistry method in 8cases of dermal ulcers and8cases of normal skins.Results In normal skins,the positive partic les of IGF-Ⅰcould be found in epidermal cells,endktheial cells a nd some fibroblasts.IGF-ⅠR was mainly located in the membranes of epidermal cells.In ulcer,IGF-Ⅰexpression was slightly enhanced but the distrbution of its positive sig nal was undifferent in comparison wi th nornal skins.IGF-ⅠRαand IGF-ⅠRβwere chiefly distrbuted in cellular membranes of some epidermal cells ,m onocyte and macrophage When compare d with normal skins,the protein expression of those two proteins was notably decreased in ulcers(P<0.01).The positive cellular ratios of two units were reduced to43.9%and 50.0%of normal skins respectively.Conclusion The formation of dermal chronic ulce rs were closely associated with the r e-duction of IGF-ⅠRαand IGF-ⅠRβprotein contents which lead to bindi ng obstruction between IGF-Ⅰand its receptor,although the invre nement of IGF-Ⅰcould hardly accelerated the recove ry of ulcers.
