黄芪注射液和白术提取部位对小肠上皮细胞移行的影响

摘　　要：目的　观察黄芪注射液 (Hi)及白术提取部位 (B1、B4、B9、B13)对小肠上皮细胞 (IEC- 6 )移行的影响 ,探讨其粘膜修复的机制。方法　 IEC- 6接种于 6孔培养板 ,72 h后损伤细胞 ,并加入不同剂量的 Hi、B1、B4、B9和 B13溶液 10 0 μL,空白组加等量 D- PBS,阳性对照组加表皮生长因子 (EGF) ,加药后 2 4 ,4 8和 72 h观察细胞移行。结果　 B4、B9、B13和 EGF均明显促进细胞移行 ,与对照组比较 2 4 ,4 8和 72 h均具有显著性差异 (P<0 .0 1) ,但B4、B9、B13作用不如 EGF强。B1和 Hi对细胞移行无明显促进作用 ,与 EGF比较 2 4 ,4 8和 72 h均具有显著性差异 (P<0 .0 1)。 B13和 Hi配伍应用后 ,对 IEC- 6细胞移行具有协同促进作用 ,与对照组、Hi和 B13组比较 2 4 ,4 8和 72 h均具有明显差异 (P<0 .0 5～ 0 .0 1)。结论　 B4、B9、B13通过促进细胞移行在小肠粘膜损伤修复过程中发挥作用 ,可能是白术粘膜保护和修复作用的物质基础。 B13和Object To observe the effects of Astragali Injection (Hi) and extracts from Atractylodes macrocephala Koidz. (B1, B4, B9, B13) on the migration of rat small intestinal crypt like cells line (IEC 6). Methods IEC 6 cells were inoculated in 6 well microplates and injured in 72 h, and added with Hi, B1, B4, B9, B13. Then D PBS to negative control in the same amount and EGF to positived control, the cell migration was observed in 24, 48 and 72 h after treatment. Results B4?B9?B13 and EGF significantly promoted the migration of wounded IEC 6 cells (P<0 01 vs the control). B1 and Hi had no effects on IEC 6 cell migration. Combined administration of B13 and Hi stimulated cell migration in a synergistic manner (P<0 05-0 01 vs the control). Conclusion B4, B9, and B13 play an important role in mucosa repair of small intestine by promoting IEC 6 cell migration, which may constitute the material basis of the protective and repairing agents induced by A. macrocephala on gastrointestinal mucosa. Compatible application of B13 and Hi shows the synergism on IEC 6 cell migration.
