摘 要:目的 研究用自体静脉移植做动脉重建后 ,血管内皮细胞早期动态变化与影响因素。方法 70只犬分成两批进行实验。第 1批实验 :40只犬按肢体远端动脉输出道不同、扩血管药物前列腺素投入与否分成 8组。第 2批实验 :30只犬按空白和抗凝、抗血小板药物肝素 ,培达的投入分成6组。移植静脉分别在术后第 1天与第 5天被取出 ,经处理后 ,在图像分析系统下测定血管内皮细胞覆盖与脱落面积。结果 术后 5d解剖检查发现静脉移植血管内皮细胞覆盖面积在肢体动脉远端输出道良好组为 85 %、肝素和培达药物投入组为 78% ,82 % ,明显高于空白组 41% (P <0 0 5 )。另外 ,无论哪一组 ,静脉移植中瓣膜部位内皮细胞覆盖面积都为 90 %以上。结论 (1)被建动脉输出道良好与否对静脉移植物血管内皮细胞的修复有明显影响。 (2 )静脉移植瓣膜部位是内皮细胞再生与修复之源 ,故做血管移植时应尽量保护。 (3)培达药物有保护静脉移植物血管内皮细胞 ,防止脱落作用。(4 )培达和肝素药物对静脉移植物血管内皮细胞的修复有明显促进作用。Objective [WT5”BZ]To study the early response of endothelial cell(EC)of the autogenous vein graft(AVG)in an arterial reconstruction model of dogs .[WT5”HZ] Methods [WT5”BZ]70 dogs were used in the experiment. 40 dogs were divided into 8 groups according to the condition of run-off of the artery that reconstructed and whether or not intravenous PGE1 was given afterwards. Then other 30 dogs were divided into 6 groups receiving respectively placebo, anticoagulant and antiplatelet, heparin and pletaal. The graft was taken out at 1st and fifth day postoperative and image analysis was used to determine the area covered by EC.[WT5”HZ]Results [WT5”BZ] It was found that the area covered by EC in the AVG was much larger in the groups with good run off (85%), receiving postoperative heparin (78%) and pletaal (82%) than the area in control group (41%, P <0 05).Moreover, the area covered by EC in the site of value of AVG was over 90% in all groups.[WT5”HZ] Conclusion [WT5”BZ] (1)Good run-off of AVG was important for the repairment of EC in the AVG. (2) The valve of the AVG was usually the origin of the nascent EC, hence should be carefully protected in the process of arterial reconstruction.(3)The medication of pletaal can protect the EC of AVG and prevent EC from falling.(4)Medication of heparin and pletaal have good effect in reendothelialization of AVG.
