摘 要:目的 探讨胃饲乙醇预处理对缺血再灌注大鼠肝脏HSP70表达的影响。方法 雄性Wistar大鼠随机分组 ,动物乙醇预处理后制作缺血再灌注模型 ,于术后 0~ 72h内不同时相活杀大鼠 ,留取血及肝脏标本保存待检。结果 实验组和对照组较正常组ALT值明显升高 ,72h后基本恢复 ,实验组恢复较快 ,但实验组和对照组间统计学差异不明显 ;病理观察见实验组较对照组表现为较轻的病理损害 ;免疫组化结果 :正常肝脏仅见弱阳性 ,散在分布于胞浆 ;术后随再灌注时相的延长 ,HSP70表达逐渐增强 ,实验组表达明显强于对照组和正常组 (P <0 0 5 ) ;WesternBlotting显示HSP70阳性条带于再灌注 3h开始增强 ,6h明显增强 ,持续至 72h达到最强 ,实验组和对照组相差显著。结论 胃饲乙醇预处理能够明显增强缺血再灌注大鼠肝脏HSP70的表达 ,从而提示与较好肝脏保护相关。Objective To investigate the expression level of HSP70 in rat liver preconditioned by ethanol gavage and its influences on the liver ischemia reperfusion injury.Methods Adult male Wistar rats were divided into different groups. Alcohol were given by gastric gavage to experimental group 24 h before 30 minute's ischemia of rat liver,and then killed at different time point between 0 to 72 h at the period of reperfusion. Blood ALT were determined and liver tissue samples were studied by immunohistochemical method, western blot and under microscope.Results 1.Pathological observation:there were obvious differences between experimental group and control group .Despite of a certain degree of inflammatory cell conglomeration at each slides,there was no obvious necrosis oberserved in experimental group as found in the control group.Immunochemistry results:hsp70(+) was found in normal liver cell plasms at a low level and increased after I/R injury. hsp70 was induced at higher level after the ischemia reperfusion injury than the control group( P <0 05).Western Blotting exmination: the hsp70 blot was enhanced at 3 h after liver reperfusion. It increased obviously 6 h after reperfusion up to 72 h. There was obvious difference between two groups.Conclusion Induced by ethanol pretreatment ,the hsp70 in rat liver can be obviously enhanced by ischemia and reperfusion injury. According to the pathological results ,it suggests that hsp70 may provide a protection to the rat liver cell.
