摘 要:目的 探讨心肌细胞内游离钙 ([Ca2 + ]i)和心肌ATP酶变化在心肌挫伤 (MC)后心功能障碍发生机制中的意义。方法 随机选取兔 3 6只 ,分为 6组 :正常对照、伤后 2、4、8、12、2 4h ,每组 6只。经右颈总动脉插管至左心室测左室压力。采用BIM Ⅱ型生物撞击机致成MC模型。在上述时相点测定左室压力 ,并测定心肌细胞内 [Ca2 + ]i和心肌匀浆组织及线粒体ATP酶活力。结果 心脏收缩 舒张功能受到损害 ,心肌细胞内 [Ca2 + ]i升高 ,心肌匀浆组织及线粒体ATP酶下降 ,相关分析表明心功能障碍与心肌细胞内 [Ca2 + ]i含量升高呈明显负相关 ,与ATP酶活性降低呈明显正相关。结论 MC后心脏功能下降 ,心肌细胞内 [Ca2 + ]i含量升高和ATP酶活性下降可能是其原因之一。Objective To explore the roles of intramyocardial calcium and myocardial ATPase in the mechanism of the cardiac dysfunction after myocardial contusion(MC)in rabbits. Methods The models of MC were established with BIM Ⅱ Bio impactor on 36 rabbits. Cardiac functions, free calcium content and the ATPase activity in myocardial tissues and cardiomyocytic mitochondria before and 0.5, 2, 4, 6, 8, 12 and 24 h after injury. Results The cardiac functions were impaired. The free calcium in myocardial cells was higher; The activities of the Ca 2+ ATPase and Na + K + ATPase in the homogenate of myocardial tissue and the cardiomyocytic mitochondria were decreased markedly after MC. There was a negative correlation between the cardiac functions with the free calcium and positive correlation between the cardiac functions with the activity of the ATPase in myocardial cells. Conclusion The cardiac functions are changed obviously after MC. The increase of the free calcium and the decrease of the activity of the ATPase in myocardial tissues and cardiomyocytic mitochondria might contribute to cardiac dysfunction after MC.
