摘 要:目的 :证实灭蚊真菌大链壶菌和卡地腐霉经诱导可产生枯草杆菌蛋白酶类蛋白酶 (Pr1)和胰蛋白酶类蛋白酶 (Pr2 )。方法 :采用特异性底物测定、X光胶片印迹试验、含底物的硝酸纤维薄膜试验等多种方法对蚊虫体壁类似物———几丁质诱导前后培养液中酶的活性进行检测和比较。结果 :大链壶菌和卡地腐霉均可产生Pr1和Pr2类蛋白酶。并且 ,经几丁质诱导 30~ 6 0h后 ,卡地腐霉产生的Pr1和Pr2类蛋白酶及大链壶菌产生的Pr2类蛋白酶活性均明显增高。并通过对培养液进行冷冻干燥、等电聚焦电泳和脱盐等处理 ,对该蛋白酶进行了初步纯化。结论 :大链壶菌和卡地腐霉均可产生Pr1和Pr2类蛋白酶。几丁质对这些类蛋白酶的释放有诱导作用。Objective:To prove whether mosquito killing fungi, Lagenidium giganteum and Pythium carolinianum, can produce Pr1 and Pr2 like proteases after induction of chitin. Methods:Special substrates test, X ray film trace test, and test with substrates treated film were adopted to search for activitie of the proteases. Results:Data showed that these fungi produced active Pr1 and Pr2 like proteases, and after 30 60 hours of induction of chintin, the activities of Pr1 and Pr2 like proteases produced by P. carolinianum and those of Pr2 like proteases produced by L. giganteum increased markedly. Primary purification of them was done with isoelectric focusing electrophoresis. Conclusion: Both L. giganteum and P. carolinianum can produce Pr1 and Pr2 like proteases, and these enzymes are inducible by mosquito cuticle like material.
