摘 要:目的 研究慢性实验应激对中国地鼠糖代谢的中枢神经内分泌机制。 方法 中国地鼠16 1只 ,分组采用三因素二水平析因设计 ,共分 8组 ,雌雄均衡匹配 ;给予震动、旋转和合并限制等实验性应激并监测血糖 ,一年后检测血浆胰岛素 (INS)、下丘脑 β-内啡肽 (β- EP)、神经肽 - Y(NPY)、促肾上腺皮质激素 (ACTH)含量。并计算胰岛素敏感指数 (ISI)。 结果 慢性实验性应激组血糖水平、下丘脑 β- EP、ACTH和 NPY含量显著高于非应激组 ,血浆胰岛素浓度和胰岛素敏感指数 (ISI)则显著低于非应激组 (P= 0 .0 0 1和 P=0 .0 2 2 )。 结论 慢性实验性应激使中国地鼠血糖增高。下丘脑 β- EP、NPY和 ACTH的含量增加 。Objective To investigate the center neuroendocrine mechanism for the effect of chronic experimental stress on plasma glucose level in Chinese Hamster. Methods According to the design of a three way ANOVA analyses, 161 three month old Chinese Hamsters were selected to assign into 8 groups and matched with sex. Experimental stressors were shaking rotation and restraint lasting one year. After 1 year we killed all rats, collected blood and hypothalamus. Then insulin (INS) in plasma, hypothalamus β endorphin (β EP), neuropeptideY (NPY), and adrenocorticotropin (ACTH) were assayed to calculate the insulin sensitivity by insulin sensitivity index [1/(BG×INS)](ISI). Results The glucose level and the β EP, NPY, ACTH concentrations in hypothalamus were significantly increased in experimental stress animals; the insulin concentrations in plasma and insulin sensitivity index were significantly lower ( P=0.001 and P =0.022) in experimental stress rats than non stress rats. Conclusion The hypothalamus β EP, NPY, ACTH concentrations are significantly influenced by stress in the onset of DM of Chinese Hamster. Therefore they might play an important role in regulating glucose metabolism in the onset of animal DM by chronic experimental stress.
