摘  要:目的:探讨前β细胞克隆增强因子(PBEF)在体外循环(CPB)术后对肺血管内皮通透性增加的调节机制。方法:对照组建立CPB后不给予CPB转流,A组给予慢病毒AD-PBEFshRNA转染,再建立CPB,不行CPB转流,B组建立CPB后给予30min深低温停循环,C组给予慢病毒AD-PBEFshRNA转染后,行30min深低温停循环。检测各组肺组织PBEF表达、磷酸化P38MAPK和磷酸化HSP27表达。结果:A、B、C组在2 h、4 h时PBEF、P38MAPK、HSP27的表达均分别明显高于0 h(P<0.01)。在2 h、4h,A、B、C组PBEF、P38MAPK、HSP27的表达均明显高于对照组,而A、B组显著低于C组,差异均有统计学意义(P<0.01)。结论:PBEF通过P38MAPK-HSP27通路增加CPB术后肺血管内皮通透性。Objective:To investigate the regulatory mechanism of Pre-B-cell colony enhancing factor increasing the permeability of pulmonary vascular endothelium after cardio pulmonary bypass.Methods:After the establishment of CPB,the control group was not treated with CPB without bypass,A group was transfected by lentivirus AD-PBEFshRNA and established CPB without bypass,B group was transfected by lentivirus AD-PBEFshRNA after the establishment of CPB and were given 30 minutes deep hypothermic circulatory arrest,C group was transfected by lentivirus AD-PBEFshRNA and were given 30 minutes deep hypothermic circulatory arrest.The expression of PBEF,phosphorylated P38MAPK and phosphorylated HSP27 in lung tissues of rats were detected.Results:The expression of PBEF,P38MAPK,HSP27 in A,B,C group in 2 h and 4 h were significantly higher that in 0 h(P<0.01).The expression of PBEF,P38MAPK,HSP27 in A,B,C group in 2 h and 4 h were significantly higher that in the control group,that of A,B group were significantly lower than those in C group(P<0.01).Conclusion:PBEF increased the permeability of pulmonary vascular endothelium through P38MAPK-HSP27 pathway after CPB.
