摘　　要：目的观察新疆阿魏乙酸乙酯分离部位对人结肠癌细胞HCT116迁移和侵袭能力的影响,探讨其相关作用机制。方法将体外不同浓度阿魏乙酸乙酯分离部位作用于人结肠癌HCT116细胞24 h,CCK-8法检测细胞增殖,计算细胞生长抑制率和IC50,细胞划痕实验和Transwell小室实验观察HCT116细胞迁移和侵袭能力。结果新疆阿魏组与顺铂组随着药物浓度的增加,OD值逐渐变小,抑制率呈药物浓度依赖性,新疆阿魏组IC50为43.98μg/m L,顺铂组IC50为36.77μg/m L。与空白组比较,经新疆阿魏树脂乙酸乙酯分离部位作用后,HCT116细胞迁移和侵袭能力明显减弱(P<0.01)。结论新疆阿魏树脂乙酸乙酯分离部位对人结肠癌细胞HCT116的增殖、迁移和侵袭能力具有一定的抑制作用。Objective To investigate the effects of separation parts of ethyl acetate from Ferula sinkiangensis resin on migration and invasion of HCT116 cells. Methods In vitro separation parts of ethyl acetate from Ferula sinkiangensis resin with different concentrations were acted on human colon cancer HCT116 cells for 24 h. CCK-8 method was used to detect cell proliferation, calculate the cell growth inhibition rate and IC50. Method of scratches the legitimacy and Transwell room were used to observe HCT116 cell migration ability. Results With the increase of medicine concentration, OD value in Ferula sinkiangensis resin group and cisplatin group decreased gradually;inhibition ratio and medicine concentration were dependent; IC50 was 43.98 μg/mL in Ferula sinkiangensis resin group and 36.77 μg/mL in cisplatin group. Compared with control group, affected by separation parts of ethyl acetate from Ferula sinkiangensis resin, migration and invasion abilities of HCT116 cells were weakened significantly (P<0.01).Conclusion Separation parts of ethyl acetate from Ferula sinkiangensis resin can reduce proliferation, migration and invasion abilities of HCT116 cells to some extent.
