“补肾健脾”针刺法对去势雌鼠OPGmRNA、ODFmRNA基因表达的影响

摘　　要：目的:观察"补肾健脾"针刺法对去势雌鼠模型骨组织OPG、ODF基因表达的影响,为该法治疗绝经后骨质疏松症提供理论依据。方法:将32只健康雌性Wistar大鼠,随机等分为4组:正常组、模型组、针刺组及雌激素组。除正常组外,对其他3组大鼠进行双侧摘卵巢手术制造绝经后骨质疏松症模型,而正常组行假手术,暴露卵巢而不摘除。正常组和模型组每日同时灌服等量生理盐水,针刺组使用36号美容针刺入双侧"足三里""肾俞""命门""关元""大杼""脾俞"穴,雌激素组用己烯雌酚溶液灌胃,3个月后取腰3锥体液氮冻存,经Trizol法抽提总RNA,RT-PCR检测OPG、ODF基因表达。结果:各组与模型组ODF m RNA表达强度相比较均极显著地降低,以雌激素组和针刺组尤为明显,而雌激素组ODF m RNA表达强度明显低于针刺组(P<0.01)。结论:补肾健脾针刺法对去势大鼠骨组织可明显抑制ODF基因的表达,促进OPG基因的表达。Objective:To observe the "Bushen Jianpi"effect of acupuncture on the expression of OPG and ODF in bone tissue of ovariectomized female rats, and to provide theoretical basis for the treatment of postmenopausal osteoporosis.Methods:32 healthy female Wistar rats were randomly divided into 4 groups:normal group,model group,acupuncture group and estrogen group.In addition to the normal group,the other three groups of rats were treated with bilateral ovarian surgery to produce postmenopausal osteoporosis model,while the normal group underwent sham operation,the exposure of the ovary and not removed.Normal group and model group were administrated saline daily at the same time, the use of acupuncture group 36,acupuncture beauty into the bilateral "Zusanli""Shenshu""Mingmen" and "Guanyuan" and "Dazhu" and "Pishu" point,estrogen diethylstilbestrol solution group by gavage,after 3 months take the waist 3 cone frozen in liquid nitrogen,total RNA was extracted by Trizol method,to detect the expression of OPG and ODF gene RT-PCR.Results:Compared with the model group,the expression intensity of ODFmRNA was significantly decreased,especially in estrogen group and acupuncture group,while the expression intensity of mRNA ODF in estrogen group was significantly lower than that in the acupuncture group(P<0.01).Conclusion:The method of invigorating the kidney and strengthening the spleen can significantly inhibit the expression of ODF gene and promote the expression of OPG gene in ovariectomized rats.
