摘  要:采用RACE技术克隆了马氏珠母贝细胞周期分裂基因45(Pm-CDC45)全长序列,利用荧光定量PCR检测该基因在闭壳肌、鳃、性腺与外套膜组织的表达量,并估计基因表达量与金黄壳色第3代选育群体壳高性状的相关性。结果表明:Pm-CDC45基因序列全长为2 091 bp,5′非编码区(5′UTR)为157 bp,3′非编码区(3′UTR)为34 bp,其中开放阅读框为1 967 bp,编码655个氨基酸;Pm-CDC45在各组织的相对表达量存在显著差异(P<0.05),其中闭壳肌表达量最高,鳃和性腺为其次,外套膜最低;Pm-CDC45基因相对表达量与选育群体壳高性状与存在显著的正相关(r=0.531,P<0.05);马氏珠母贝Pm-CDC45为影响壳高性状基因。The full length of Pm-CDC45 was obtained by using rapid amplification of cDNA endstechnology. Gene expression was detected in adductor muscle,gill,gonad and mantle. Correlationcoefficient was estimated between gene expression and shell height of the third generation yellowcolored selected line. The results showed that the total length of Pm-CDC45 gene length was 2 091 bp,including 1 967 bp of the open reading frame (ORF) which encodes 655 amino acids, a 5′ UTR of 157bp and a 3′ UTR of 34 bp. There existed significant differences in relative expression of Pm-CDC45among the four tissues (P < 0.05). The highest gene relative expression was observed in adductormuscle, followed by gill, gonad and mantle. There existed a significantly positive relationship between gene relative expression and shell height (r = 0.531, P < 0.05). The present results suggest thatPm-CDC45 is an important gene that affects shell height in pearl oyster P. fuctada martensii.
