摘 要:为探讨虎斑乌贼受精卵最佳孵化条件,根据中心复合试验设计,在单因子试验基础上,用响应曲面法分析温度(24~32℃)与pH(7.0~8.5)对受精卵孵化率的联合效应,获得温度与pH的最佳组合。结果表明:在本试验条件下,温度和pH的一次、二次效应及两者的互作效应对孵化率影响均有统计学意义(P<0.01);随着温度或pH的上升,孵化率呈先升后降的变化趋势,对数据进行二次多元回归拟合,得孵化率(Y)对实际自变量温度(X1)和pH(X2)的二次回归方程.=85.27-6.74X1+5.22X2+9.42X12-12.43X12-8.18X22,回归方程的相关指数为0.9786,校正系数为0.9633,精密度达22.974,可用于预测与分析。回归方程优化显示,温度为27.5℃和pH7.9时,孵化率最大值为86.46%,模型的可信度高达0.949。验证结果表明,该组合条件与预测值相符。On the basis of one-factor tests and the central composite design (CCD) principle, the method ofresponse surface analysis was adopted to evaluate the combined effects of temperature (24–32 ℃) and pH(7.0–8.5) on hatchability of Sepia pharaonis. The results showed that the linear and quadratic effects oftemperature and pH had a very significant effect on hatchability (P < 0.01).The interactive effect betweentemperature and pH also had a very significant effect (P < 0.01). Hatchability increased and then decreasedas the temperature or pH increased. The model equation of hatchability (Y) towards temperature(X1) and pH(X2) were established as follows: ? = 85.27–6.74 X1 + 5.22 X2 + 9.42 X12–12.43 X12–8.18 X22 (R2 = 0.978 6;adjusted R2 = 0.963 3; Adeq precision = 22.974). The experimental results indicated that the proposed modelwas reasonably accurate and can be used for describing and predicting the hatchability. Solving theregression equation for temperature equals to 27.5 ℃ and pH equals to 7.9, the optimum predicted valuefor hatchability was 86.46% and the reliability peaked to 0.949. Moreover, since verification result ofoptimum conditions accorded with the predicted value, it is suggested that the hatching of fertilized egg of S.pharaonis can be improved by rearing at an optimal combination of temperature and pH.
