摘  要:目的:观察地佐辛预先给药对切口痛大鼠中脑导水管周围灰质c-fos蛋白表达的影响。方法:成年雄性Wistar大鼠,36只,随机分为3组(n=12),对照组(C组)仅给予麻醉而不手术;生理盐水组(S组):切口疼痛模型手术前15 min尾静脉注射0.9%氯化钠溶液2 ml;地佐辛组(D组):切口疼痛模型手术前15 min尾静脉注射地佐辛3 mg/kg 2 ml。于术前24 h(T_0)、术后2 h、6 h、24 h(T_(1-3))时测定机械缩足反射阈值(mechanical withdrawal threshold,MWT)和累积疼痛评分,T3时痛阈测定后处死大鼠,取大鼠全脑,采用免疫组化方法检测中脑导水管周围灰质神经元c-fos蛋白表达。结果:与C组相比较,S组和D组T1-3时MWT明显降低,累积疼痛评分升高,中脑导水管周围灰质神经元c-fos蛋白免疫阳性细胞数增高(P<0.05);与S组比较,D组T1-3时MWT升高,累积疼痛评分降低,中脑导水管周围灰质神经元c-fos蛋白免疫阳性细胞数下降(P<0.01)。结论:地佐辛预先给药可抑制中脑导水管周围灰质神经元c-fos蛋白表达,减轻大鼠切口痛。Objective: To observe the effect of dezocine pretreatment on c-fos protein expression in periaqueductal gray of rats with incisional pain. Methods: Thirty-six male Wistar rats were randomly divided into 3 groups (n =12 each): control group(group C), normal saline group (group S), and dezocine group (group D).In the saline or dezocine group, rats were injected with normal saline 2 ml or dezocine 3 mg/kg (in 2 ml of volume) via tail vein. Rats in group S and group D received surgery to establish incisional pain model 15 min after tail vein injection. The mechanical paw withdrawal threshold (MWT) and cumulative pain score were measured 24 h before surgery (T0, baseline) and 2 h, 6 h, 24 h after surgery (T1-3). The rats were sacrificed after all of the pain threshold measurement and the whole brains were collected for detecting the c-fos expression in periaqueductal gray by immunohistochemistry assay. Results: In group S and group D, the MWTs were significantly decreased, cumulative pain scores were significantly increased, and the expression of c-fos protein was significantly up-regulated at T1-3, compared with group C. Compared with group S,the rats in group D exhibited increased MWT, decreased cumulative pain score, and down-regulated c-fos expression at T1-3. Conclusion: Dezocine can inhibit the up-regulation of c-fos protein expression and thus attenuate the incisional pain in rats.
