摘 要:目的通过观察青蒿琥酯(Art)对高糖培养条件下大鼠肾小球系膜细胞(HBZY-1)的增殖、凋亡及核因子(NF)-κB、单核细胞趋化蛋白-1(MCP-1)表达的变化,探讨Art对糖尿病肾病保护性作用的机制。方法传代培养大鼠肾小球系膜细胞HBZY-1,分为低糖(5.6 mmol/L葡萄糖)组、高糖(25 mmol/L葡萄糖)组、高糖+不同浓度青蒿琥酯(10、20、30 mg/L)组。48 h后采用四甲基偶氮唑盐微量比色(MTT)法观察细胞增殖能力改变;流式细胞仪Annexin V-FITC/PI双染法检测细胞凋亡指数;酶联免疫吸附试验(ELISA)法检测细胞上清液NF-κB、MCP-1的含量。结果 (1)经高糖处理48 h后,HBZY-1细胞较低糖组增殖能力及凋亡率均增加,细胞上清液NF-κB、MCP-1浓度升高。(2)经Art干预后,与高糖组比较,HBZY-1细胞增殖抑制,细胞凋亡率增加,细胞上清液NF-κB、MCP-1浓度降低,其干预作用呈现剂量依赖性(P<0.05)。结论 Art能剂量依赖性地逆转高糖的作用。Art对高糖诱导下肾小球系膜细胞的增殖与凋亡以及炎症因子的作用机制可能为临床上糖尿病肾病提供新的治疗策略。Objective To investigate the effects of artesunate(Art)on cell proliferation,apoptosis,nuclear factor(NF)-κB and monocyte chemotactic protein1(MCP-1)expressions induced by high glucose in rat renal mesangial cells(HBZY-1),and the mechanism thereof.Methods HBZY-1cells were cultured and divided into normal glucose group(5.6mmol/L),high glucose group(25mmol/L)and high glucose with different concentrations of Art(10mg/L,20mg/L,30mg/L)groups.MTT assay was used to detect the cell proliferation after48h.The apoptotic rate was evaluated by flow cytometry withAnnexin V-FITC/PI double stains.The protein levels of NF-κB and MCP-1in the cell culture supernatant were determinedusing ELISA.Results High glucose induced apoptosis and proliferation in HBZY-1cells,and the expressions of NF-κBand MCP-1in the supernatant were also increased(P<0.05).After treatment with Art,the proliferation was obviouslyabolished,and the apoptosis was increased,and the expressions of NF-κB and MCP-1in the supernatant were decreased inHBZY-1cells.The effects of Art showed a dose-dependent manner(P<0.05).Conclusion Artesunate treatment canreverse the effect of high glucose in HBZY-1cells in a dose-dependent manner,which may provide a new therapeuticstrategy for diabetic nephropathy.
