摘  要:目的观察鹿红方对心力衰竭大鼠过氧化物酶增殖物激活受体(PPAR)和心肌组织的影响。方法分离鹿红方全方水煎液,用蒸馏水、30%乙醇、60%乙醇和95%乙醇洗脱,得到4种提取液,分别成为A、B、C、D液,分组为A组、B组、AB组、C组、AC组、BC组、ABC组、D组、AD组、BD组、ABD组、CD组、ACD组、BCD组,共14组,第15组曲美他嗪,第16组假手术组。分别给96只通过冠脉结扎法制备充血性心力衰竭成功的大鼠模型灌胃。应用Western Blot检测(WB)测定不同剂量的鹿红方对心肌细胞PPARα蛋白表达的影响,对心肌组织进行HE、VG染色,观察心肌细胞病理形态结构。结果鹿红方具有明显上调PPARa的作用,可以让无序的心肌纤维定向排列,使心衰心脏的心功能得以改善,对心衰病理改变有显著降胶原纤维的作用,有显著降低PVCA的功效。结论不同剂型鹿红方药物血清组都可通过上调PPARα的表达,发挥其改善心肌组织功能作用,表明鹿红方是中医药治疗心力衰竭的良好药物。Objective To observe the effects of Luhong prescription (LHP)on peroxisome proliferator activated receptor (PPAR) and myocardium of mouse with congestive heart failure (CHF).Methods Separation water decocted solution of LHP was eluted with distilled water,30% ethanol and 60% ethanol,95% ethanol,extract,respectively,as a,B,C,D solution,grouped into A,B and AB,C,AC,BC and ABC,D,ad and BD,abd,CD,ACD,BCD in a total of 14 groups,group 15 (trimetazidine)and group 16 (sham operation group),respectively.Ninety six rat models of CHF were established by coronary artery ligation and administrated by intragastric ad- ministration.The protein expression of PPARαon cardiomyocyte was detected by western blot (WB).The pathological structure of my- ocardial tissue was observed by Haematoxylin Eosin (HE)staining and Van Gieson (VG)staining.Results LuHong prescription can upregulate the function of PPARa obviously,it can make the muscle fiber orientation disordered,it can make the heart to improvethe heart function of chronic heart failure.LuHong prescription can reduce collagen on the pathological changes of heart failure. Lu- Hong prescription can reduce the efficacy of PVCA significantly.Conclusion LHP can upregulate the protein expression of PPARα on cardiomyocyte in rats with CHF.
