摘 要:目的通过观察地塞米松诱导C57BL/6小鼠脂肪和肌肉的变化,探究建立理想少肌性肥胖(sarcopenic obesity,SO)小鼠模型的方法。方法 23只6~7月龄雄性C57BL/6小鼠按随机数字表法分为对照组(n=11)和实验组(n=12)。对照组采用0.9%生理盐水、实验组采用10 mg/kg地塞米松磷酸钠注射液,连续皮下注射6周,分别于造模第3、6、9、12、15、21、28、35、42天测量两组的摄食量、体重和体成分;最后一次给药24 h后,利用水迷宫测量小鼠的游泳速度和轮式跑台测量小鼠的掉落次数以评价小鼠的肌肉功能。结果(1)从造模第1天开始,实验组平均每日摄食量较对照组明显增加[(4.49±0.22)g vs(3.55±0.21)g;t=-8.554,P<0.05];(2)从造模第21天起,实验组平均体重较对照组明显增加[(34.22±2.28)g vs(31.03±2.65)g;t=-3..345,P=0.003](3)体成分方面,从造模第28天起,实验组平均筋肉含量较对照组明显降低[(21.55±1.21)g vs(23.23±1.40)g;t=3.657,P=0.001],而从造模第6天起,实验组平均脂肪含量较对照组明显增加[(8.80±1.5 8)g vs(4.96±1.40)g;t=-5.773,P<0.05];(4)肌肉功能方面,实验组平均游泳速度较对照组显著下降(t=2.825,P=0.010),平均轮式跑台掉落次数较对照组显著增多(t=-2.094,P=0.020)。结论连续皮下注射地塞米松诱导成年小鼠能够建立理想的SO小鼠模型。Objective The study objective was to explore the construction method of an ideal sarcopenic obesity (SO) mouse model by observing the changes of fat and muscle in C57BL/6 mice induced by dexamethasone. Methods Twenty-three 6-7 months old male C57BL/6 mice were divided into control group (n=11) and experimental group (n=12) by the means of random number table allocation, the control group was treated with 0.9% normal saline while the experimental group was treated with 10 mg/kg dexamethasone sodium phosphate injection, both groups were given subcutaneous injections for consecutive six weeks. The food intake, body weight and body composition of these mice were measured every three days in the first two weeks and every seven days in the following four weeks; to evaluate the function of the mice comprehensively the swimming speed in the water maze and the number of times the mice fell off the rotary treadmill 24 hours after the last dosing were measured. Results ( 1) From the first day of the model establishment, the average daily food intake of the experimental group increased significantly compared to the control group [(4.49±0.22) g vs (3.55±0.21) g;t=-8.554,P<0.05]; (2) From the twenty-first day of the model establishment, the average weight of the experimental group increased significantly compared to the control group [(34.22±2.28)g vs (31.03±2.65) g; t=-3.345, P=0.003]; (3) As for the body composition, from the twenty-eighth day of the model establishment, the average muscle content in the experimental group was significantly lower than the control group [(21.55±1.21) g vs (23.23±1.40) g; t=3.657, P=0.001], and from the sixth day of the model establishment, the average fat content in the experimental group was significantly increased compared to the control group [(8.80±1.58) g vs (4.96±1.40) g; t=-5.773, P<0.05];(4) As for muscle function, the average swimming speed of experimental group decreased significantly compared to the control group (t=2.825,P=0.010), the average number of times the experiment
