摘 要:为了从废弃烟叶高效提取烟碱,本文利用响应面法对在废弃烟叶中烟碱的提取条件进行优化。以烟碱的得率为评价指标,设计酶解时间、酶解温度,p H值及提取时间4因素3水平的实验模型,采用Design Expert 8.0.6分析软件对烟碱的提取条件进行优化研究。优选出最佳提取工艺为:酶解时间2.0 h,酶解温度48.50℃,酶用量2.10%,p H4.00。测定烟碱的得率为3.320%,与预测值3.32836%偏差较小,为废弃烟叶的烟碱提取提供理论依据。To optimize the conditions for purification of Nicotine in abandoned Tobacco.Response surface method was used to design the test models with four factors including enzyme time,enzyme temperature,enzyme dosage,ultrasonic time at three levels based on the single factor experiment and the yield of Nicotine was used as the evaluation index to optimize purification process of Nicotine.The optimum extraction condition were as follows:enzymolysis time was2.00h,enzymolysis temperature was48.50℃,cellulose enzyme amount was2.10%,pH was4.00.Under this condition,the yield was up to3.320%,the relative error was smaller compared to the predicted value3.32836%.The optimum extraction conditions were simple and stable and they were served as a reference for industrial large scale production.
