摘  要:基于次溴酸盐氧化铵盐(NH+4)成为亚硝酸盐(NO2~–)的原理,发展了一种小体系连续快速测定海洋水体溶解性铵盐的方法。该方法以圆底旋盖2 mL离心管为反应容器,应用次溴酸钠氧化剂氧化铵离子成为亚硝酸根离子,进一步通过加入Griess试剂生成有色物质,然后利用标准酶标板为底板,采用酶标仪对生成的有色物质进行吸光值测定。此方法空白吸光值低,标准曲线在0—32μmol·L^(-1)浓度范围内线性良好(R^2>0.99),可批量连续测定样品。对环境样品的测定结果表明,小体系铵盐测定法具有较高的精确度,与《海洋规范》中标准25 mL体系法测定的数值具有大于99%的拟合度,为海洋水体NH+4-N的实地测定提供了一种快速有效的新方法。Based on the chemical reaction of oxidation of NH+4into NO2-by bromite oxidant,a fast and sequential method was devolved in small volume.In this method,a2mL Oak Ridge centrifuge tube was selected as reaction vessel,and the dissolved NH+4was converted to NO2-with bromite as oxidant.The generated NO2-was determined with Griess reagent by a Microplate Reader.The absorption blank was relatively low,and the calibration curve had rigid linear regression(R2>0.99)in the range of0-32μmol·L?1.Environmental samples assay demonstrated that this small volume method was well consistent with conventional method(in25mL volume)for ammonia determination.In total,this modified method improved accuracy and efficiency of operations greatly,and would be realized as a rapid and high-throughput determination of ammonia in field analysis with low cost.
