摘  要:目的:研究三草汤总黄酮对佐剂性关节炎大鼠的保护作用及可能的机制。方法:将40只健康雄性SD大鼠随机分为溶媒对照组(C组)、模型组(M组)、三草汤总黄酮低剂量给药组(L组)、三草汤总黄酮高剂量给药组(H组),每组10只。用弗式完全佐剂建立大鼠AA模型,建模前1h(D0)给予三草汤总黄酮,而后由建模后第1天开始,每天灌胃1次至建模后第9天,连续灌胃给药共10天(D0~9),模型组和对照组大鼠给予等体积生理盐水。用ELISA法检测血清TNF-α、IL-1和IL-6的水平。HE染色法观察炎症关节病理学变化,检测足容积变化及大鼠关节软骨组织ASIC1a蛋白表达的变化。结果:与模型组相比,给药组大鼠的右后足红肿消失明显加快。给药组能够明显降低大鼠血清IL-6、IL-1的含量且下调大鼠关节软骨组织ASIC1a蛋白的表达,对血清TNF-α的含量没有影响。结论:三草汤总黄酮对大鼠佐剂性关节炎具有保护作用,其作用机制可能与减少炎症介质IL-6、IL-1的释放及抑制关节软骨组织ASIC1a蛋白表达有关。Objective:To study the protective effect of total flavonoids in the Sancao decoction on rats with adjuvant arthritis and the possible mechanism.Methods:40healthy male SD rats were randomly and equally divided into the control group(group C),Model group(group M),low dose of total flavonoids group(group L)and high dose of total flavonoids group(group H).The model of AA rat was established by FCA.Total flavonoids in the Sancao decoction were given rats1h(D0)before the model was established.After the model was established,intragastric administration was carried out every day until the ninth day.Group C and group M were given the same volume of normal saline.The levels of serum TNF-α,IL-1and IL-6were detected by ELISA.HE staining was used to observe the pathological changes of inflammatory joints.The changes of foot volume and the expression of ASIC1a protein in articular cartilage of rats were detected.Results:Compared with group M,right rear foot swelling of rats in group L and group H was disappeared significantly.What's more,the level of serum IL-1and IL-6was significantly reduced under the action of total flavonoids in the Sancao decoction;and the expression of ASIC1a protein in rat articular cartilage tissue was down-regulated.However,there is no effect on the level of serum TNF-α.Conclustion:Total flavonoids in the Sancao decoction showed protective effects on rat AA;the mechanism may be related to reducing the release of inflammatory mediators of IL-6,IL-1and inhibiting the expression of ASIC1a protein in articular cartilage tissue.
