摘 要:目的探讨灯盏花素对糖尿病心肌病大鼠微血管基底膜及肌组织胶原容积分数(CVF)的影响。方法 SD大鼠40只,随机分为对照组(NC组)、糖尿病心肌病组(DCM组)、灯盏花素组(Bre组),实验末测定各组VG胶原染色图像分析仪测量心肌胶原容积分数;电镜下对比各组大鼠心肌超微结构。结果 NC组毛细血管基底膜厚度为(114.61±6.80)nm,CVF值为(9.2±1.9)%;DCM组毛细血管基底膜厚度为(219.27±13.98)nm,CVF值为(20.6±3.3)%;Bre组毛细血管基底膜厚度为(196.85±10.11)nm,CVF值为(17.8±2.1)%;与NC组比较,DCM组及Bre组其基底膜厚显著降低,差异具有统计学意义(P均<0.01);Bre组基底膜厚明显低于DCM组,差异具有统计学意义(P<0.05)。与NC组比较,DCM组及Bre组CVF值明显升高,差异具有统计学意义(P均<0.01),与DCM组比较,Bre组CVF值降低,差异具有统计学意义(P<0.05)。结论灯盏花素可以改善受微血管病变,抑制了糖尿病大鼠心肌进一步纤维化。Objective To investigate influence by breviscapine on microvascular basement membrane and muscle tissue collagen volume fraction(CVF)in mice with diabetic cardiomyopathy.Methods A total of40SD rats were randomly divided into control group(NC group),diabetic cardiomyopathy group(DCM group)and breviscapine group(Bre group).Cardiac muscle tissue collagen volume fraction in all groups were detected by VG collagen staining images analyzer at the end of the experiment,and myocardial ultrastructure were compared by electron microscope.Results NC group had capillary basement membrane thickness as(114.61±6.80)mm and CVF as(9.2±1.9)%.DCM group had capillary basement membrane thickness as(219.27±13.98)mm and CVF as(20.6±3.3)%.Bre group had capillary basement membrane thickness as(196.85±10.11)mm and CVF as(17.8±2.1)%.DCM group and Bre group had obviously lower capillary basement membrane thickness than NC group,and their difference had statistical significance(all P<0.01).Bre group had much lower capillary basement membrane thickness than DCM group,and the difference had statistical significance(P<0.05).DCM group and Bre group had much higher CVF than NC group,and the difference had statistical significance(all P<0.01).Bre group had lower CVF than DCM group,and their difference had statistical significance(P<0.05).Conclusion Breviscapine can improve microangiopathy and inhibit further myocardial fibrosis in rats with diabetic cardiomyopathy.
