摘  要:目的探讨辽东楤木叶总皂苷(ETS)诱导人结肠癌HT-29细胞凋亡及其对凋亡相关蛋白表达的影响。方法 MTT法检测不同浓度(6.25、12.5、25、50、100、200 mg/kg)ETS培养的结肠癌HT-29细胞增殖率,Hoechst33258染色和激光共聚焦成像检测细胞凋亡,并观察凋亡细胞的形态学改变,免疫组化检测凋亡相关蛋白Bcl-2、Bax的表达。结果 ETS可诱导HT-29细胞发生凋亡,且凋亡作用在一定范围内呈剂量依赖性;ETS可使HT-29细胞Bcl-2表达降低、Bax表达升高(P<0.05,P<0.01),且呈明显的量效关系。结论 ETS可诱导HT-29细胞凋亡,其凋亡机制可能与降低Bcl-2、升高Bax的表达相关。Objective To study the apoptosis of human colorectal cancer HT-29cells induced by Aralia elataSeem leaf total saponin(ETS)and its effects on the expression of relevant proteins.Methods MTT assay was used todetect the proliferation of human colorectal cancer HT-29cells cultivated with different concentrations(6.25,12.5,25,50,100,200mg/kg)of ETS.Hoechst33258staining and laser confocal imaging were used to detect the apoptoticcells.Morphological changes were observed.The expressions of Bcl-2and Bax were detected by immunohistochemistry.Results ETS could induce apoptosis of HT-29cells and apoptosis was in a dose-dependent manner ina certain range.ETS could decrease the expression of Bcl-2and increase the expression of Bax in HT-29cells(P<0.05,P<0.01),showing a significant dose-effect relationship.Conclusion ETS can induce the apoptosis of HT-29cells,and the mechanism may be related to reducing the expression of Bcl-2and increasing the expression of Bax.
