针刺对血管性痴呆大鼠前额叶皮层氧化应激及神经元损伤的影响

摘　　要：目的观察针刺对慢性脑低灌注大鼠前额叶皮层氧化应激及神经元损伤的影响,探讨针刺改善血管性痴呆(VD)大鼠认知损害的分子机制。方法采用永久性双侧颈动脉结扎法造成大鼠慢性脑低灌注复制VD模型,造模后大鼠随机分为模型组、针刺组和非穴组,同时设假手术组作为对照。针刺组和非穴组造模后3 d针刺治疗,2周后灌注取材。尼氏染色检测前额叶皮层神经元数量,DHE染色检测活性氧(ROS)含量,黄嘌呤氧化酶法检测超氧化物歧化酶(SOD)活性。结果模型组神经元数量、SOD活性较假手术组降低,ROS含量升高;针刺组神经元数量和SOD活性较模型组明显升高、ROS含量降低,同时非穴组各指标未见明显改善。结论针刺可抑制VD大鼠前额叶皮层的氧化应激水平,改善神经元损伤和丢失。Objective To observe the effects of acupuncture on oxidative stress and neuronal damage in theprefrontal cortex of chronic cerebral hypoperfusion(CCH)rats;To investigate the molecular mechanisms ofacupuncture in cognitive impairment of vascular dementia(VD)rats.Methods The animal model of VD wasreplicated by permanent bilateral common carotid artery occlusion(2VO)to induce CCH in rats.After modeling,ratswere randomized into model group,acupuncture group and non-acupoint group.Sham-operation group was set ascontrol group.Rats of acupuncture group and non-acupoint group were treated with needling for2weeks at3daysafter2VO.Nissl and DHE staining were respectively used to evaluate the numbers of neurons and ROS level.SODactivity was examined by xanthine oxidase technique.Results Compared with sham-operation group,the number ofneurons and SOD activity decreased in the model group,while the ROS level increased.Compared with the modelgroup,the number of neurons and SOD activity increased in the acupuncture group,while the ROS level decreased.There were no obvious changes in all indexes in non-acupoint group.Conclusion Acupuncture can inhibit oxidativestress and improve neuronal damage and loss in the prefrontal cortex of VD rats.
