黄芪-当归不同配伍比例对大鼠血管内膜增生的影响

摘　　要：目的观察黄芪和当归不同比例配伍对大鼠血管内膜增生的影响,确立二者抗血管内膜增生的有效配伍。方法采用基线等比设计方法将SD大鼠分为5个黄芪-当归不同配比组、单用黄芪组、单用当归组、阳性药组和假手术组,采用球囊导管损伤大鼠血管内皮造成胸腹主动脉血管内膜增生模型,同时灌胃黄芪、当归不同比例配伍药物,连续14 d,放血处死大鼠,取胸腹主动脉,行Masson染色,形态计量学方法分析血管内膜增生情况。结果血管内膜损伤后14 d,胸腹主动脉内膜面积(IA)、内膜厚度(IT)、内膜面积增生率(HRIA)、内膜厚度增生率(HRIT)均增加。与模型组比较,单用当归组、芪归1∶2组、芪归1∶5组、芪归1∶1组、芪归5∶1组IA、IT、HRIA、HRIT均降低,且以芪归1∶1组作用最强。单用黄芪组、芪归2∶1组内膜增生各指标变化不明显。结论黄芪和当归在一定比例范围配伍时具有抑制血管内膜增生的作用,其中以黄芪∶当归=1∶1配伍比例抗血管内膜增生的作用为佳。Objective To observe the effects of different compatibility ratios of Astragali Radix and AngelicaeSinensis Radix on vascular intimal hyperplasia;To ascertain the effective compatibility of Astragali Radix andAngelicae Sinensis Radix for antagonizing vascular intimal hyperplasia.Methods SD rats were divided into differentgroups by baseline geometric design method:Astragali Radix and Angelicae Sinensis Radix different compatibilityratios groups,Astragali Radix group,Angelicae Sinensis Radixgroup,positive medicinegroup and sham-operationgroup.A model of intimal hyperplasia of thoracoabdominal aorta was established by balloon catheter injury invascular endothelium of rats.Then the thoracoabdominal aorta was taken out after gavage of Astragali Radix andAngelicae Sinensis Radix with different compatibility ratios for14days.Bloodletting was used to takethoracoabdominal aorta.Masson staining and Morphometric methods were used to analyze the intimal hyperplasia.Results IA,IT,HRIA and HRIT increased14day after intimal injury.Compared with the model group,IA,IT,HRIAand HRIT in Angelicae Sinensis Radix group,Astragali Radix and Angelicae Sinensis Radix1:2group,AstragaliRadix and Angelicae Sinensis Radix1:5group,Astragali Radix and Angelicae Sinensis Radix1:1group and AstragaliRadix and Angelicae Sinensis Radix5:1group were lower,and the effects of Astragali Radix and Angelicae SinensisRadix1:1ratio were strongest.The effects on intimal hyperplasia in Astragali Radix group and Astragali Radix and Angelicae Sinensis2:1group had no significant differences compared with model group.Conclusion Astragali Radixand Angelicae Sinensis can inhibit vascular intimal hyperplasia in a certain compatibility ratios,and the effects ofAstragali Radix and Angelicae Sinensis Radix1:1on intimal hyperplasia are the best.
