摘 要:目的:探讨铁超载对非酒精性脂肪肝病(NAFLD)模型大鼠脂代谢的影响。方法:48只雄性SD大鼠随机分成空白对照组(基础饲料)、高铁组(含1%FeSO_4的基础饲料)、高脂组(高脂饲料)、高脂低铁组(含0.5%FeSO_4的高脂饲料)、高脂高铁组(含1%FeSO_4的高脂饲料)。干预20周后,称取大鼠体质量;肝脏油红O染色观察肝脏脂质堆积情况;测定肝脏内甘油三酯(TG)含量;实时荧光定量PCR(q PCR)和Western blot分别检测大鼠肝脏中脂肪酸合成酶(FAS)、肉碱酰基转位酶Ⅰ(CPT1)的mRNA和蛋白表达水平。结果:与对照组比较,高脂膳食20周使大鼠体质量、肝脏TG含量均明显增加(P均<0.05),并促进TG在肝脏的沉积;且FAS的mRNA和蛋白水平均增加,CPT1的mRNA和蛋白水平均降低(P均<0.05)。与高脂组比较,高脂高铁组大鼠肝脏TG含量明显增加(P<0.05);FAS的mRNA和蛋白水平均显著增加,CPT1的mRNA和蛋白水平均降低(P均<0.05)。结论:铁超载能够促进NAFLD大鼠肝脏脂质堆积,加重脂代谢紊乱,促进NAFLD病程进展。OBJECTIVE:To explore the effects of high fat combined with iron overload diet in rats on lipid metabolism in the development of nonalcoholic fatty liver disease(NAFLD).METHODS:Forty-eight male SD rats were randomly divided into5groups:normal control,high iron diet(high iron diet containing1%FeSO),high fat diet(high4fat diet),high fat and low dose iron supplement diet(high fat diet containing0.5%FeSO),high fat and high dosage iron4diet(high fat diet containing1%FeSO)groups.These rats were sacrificed after20weeks.Their body weights were4measured and liver lipid accumulation was observed by oil red O staining.The content of liver TG was measured.Quantitative real-time PCR(qPCR)were used to detect mRNA levels of fatty acid synthase(FAS)and carnitine palmitoyl transferase1(CPT1)and their proteins were detected by Western blot.RESULTS:Compared with the normal control group,the body weight and liver TG content in the high fat group were increased significantly(P<0.05)and high fat diet promoted the deposition of TG in the liver.Additionally,the mRNA level and protein expression of FAS were significantly increased,but the mRNA level and protein expression of CPT1were significantly decreased(P<0.05).Compared with the high fat group,the content of liver TG in the high fat and high dosage iron supplement group was significantly increased(P<0.05).The mRNA and protein level of FAS were increased significantly and the mRNA and protein level of CPT1were significantly decreased(P<0.05).CONCLUSION:Iron overload further promoted the deposition of TG in the liver,the disorder of lipid metabolism and the development of NAFLD.
