金钗石斛对糖尿病大鼠肝脏TNF-α表达的影响

摘　　要：目的:观察金钗石斛对糖尿病大鼠肝脏TNF-α表达的影响。方法:高糖高脂饲料联合链脲佐菌素建立糖尿病大鼠模型。药物干预6周后,测定血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST),取肝脏称重,计算肝脏指数,用ELISA法和Real-time PCR法分别测定肝组织肿瘤坏死因子α(TNF-α)的含量和表达。结果:与模型组相比,金钗石斛组血清ALT和AST活性显著降低,肝脏指数下降,肝组织中TNF-α的含量和表达均下降了17.41%和38.01%(P<0.05或P<0.01)。结论:金钗石斛对糖尿病大鼠肝脏具有保护作用,其机制可能与下调TNF-α的表达有关。Objective:To observe the effect of Jinchai Shihu on the expression of TNF-αin liver of diabetic rats.Methods:Model ofdiabetic rats were established by high glucose and high fat diet with streptozotocin.The serum levels of ALT and AST were measured afterintervention for6weeks.Liver index was calculated.TNF-αwas determined by ELISA and Real-time PCR.Results:Compared with themodel group,the serum ALT and AST activities of the Jinchai Shihu group were significantly decreased and the liver index were decreased,and the content and expression of TNF-αin liver tissue were decreased by17.41%and38.01%(P<0.05or P<0.01).Conclusion:JinchaiShihu has protective effects on liver of diabetic rats,and its mechanism may be related to the down-regulation of TNF-αexpression.
