摘 要:目的探讨3,3′-二吲哚甲烷(3,3′-diindolylmethane,DIM)对2 Gy辐射损伤小鼠造血系统的保护作用。方法 30只雄性C57BL/6小鼠随机区组法分为对照组、2 Gy照射组和2 Gy照射+DIM组,每组10只。对照组小鼠接受假照射(0 Gy),其余2组小鼠接受2 Gy^(137)Csγ-射线一次性全身照射。2 Gy照射+DIM组小鼠照射前30 min腹腔注射DIM(75 mg/kg),其余2组小鼠腹腔注射对照溶液。照后7 d和15 d每组取5只小鼠,取外周血进行血象测定,取骨髓细胞测定骨髓有核细胞(BMNCs)数目,活性氧检测探针(DCFH-DA)检测细胞内活性氧(ROS)水平、骨髓粒-单核巨噬细胞集落(CFU-GM)形成数目检测克隆形成能力。结果与对照组比较,2 Gy照射组外周血白细胞、血小板数量、BMNCs数目和克隆形成能力显著下降,细胞内ROS水平显著提高(P<0.05)。与2 Gy照射组比较,2 Gy照射+DIM组外周血白细胞、血小板数量、BMNCs数目和克隆形成能力显著提高,细胞内ROS水平显著下降(P<0.05)。结论 DIM对小鼠2 Gy照射后造血系统损伤有一定的保护作用,其机制可能与降低骨髓细胞内ROS水平有关。Objective To investigate the protective effect of3,3′-diindolylmethane(DIM)on radiation-induced injuryin mouse hematopoietic system.Methods Thirty C57BL/6mice were randomly divided into control group,2Gy irradiationgroup and2Gy irradiation+DIM group(n=10for each group).Mice of control group received sham irradiation,and the othertwo groups accepted2Gy137Csγ-ray total body irradiation.Mice in2Gy irradiation+DIM group were intraperitoneallyinjected75mg/kg DIM30min before irradiation.Mice of other two groups were treated with reference solution.After7d and15d of2Gy irradiation,the peripheral blood samples were collected to count the number of bone marrow nuclear cells(BMNCs).The level of reactive oxygen species(ROS)was measured by DCFH-DA.The levels of colony forming unitsgranulocyte-macrophage(CFU-GM)were also detected.Results The numbers of white blood cell(WBC),platelet count(PLT),BMNCs and CFU-GM were significantly decreased and the ROS level of bone marrow cells increased significantly inthe irradiated group than those of control group(P<0.05).Compared to2Gy irradiation group,the numbers of WBC,PLT,BMNCs and CFU-GM were significantly increased in2Gy irradiation+DIM group,and the level of ROS was decreasedsignificantly(P<0.05).Conclusion DIM has a protective effect on hematopoietic cells following radiation-induced injury,which may be related with the decreased ROS level.
