摘 要:目的研究并明确肿瘤细胞膜纳米粒载药系统的体外应用价值。方法本实验设计并成功构建了一种新型肿瘤细胞膜纳米粒载药系统。该系统以肿瘤细胞膜为载体,以聚乳酸-羟基乙酸共聚物(PLGA)为内核,负载香豆素-6(Coumarin-6)作为荧光探针。在体外通过流式细胞仪及激光共聚焦成像分析并评价肿瘤细胞膜纳米粒(CNPs)及红细胞膜纳米粒(RNPs)、脂质体纳米粒(LNPs)在肿瘤细胞中的摄取差异。结果 CNPs可粘附到肿瘤细胞表面,且肿瘤细胞对CNPs摄取最多。结论肿瘤细胞膜纳米粒载药系统在肿瘤靶向递药领域具有应用前景。Objective For investigating and assessing the application of cancer cell membrane coated nanoparticles(CNPs).Methods In thisresearch,we designed a innovative nanoparticle with cancer cell membrane coating and coumarin-6labled PLGA core.We evaluated the uptake betweenCNPs and non-CNPs(including red blood cell membrane coated nanoparticles and liposome nanoparticles)of cancer cells in vitro,by flow cytometryanalysis and fluorescently confocal imaging.Results Through data analysis,we found that CNPs could specially adhere to the surface ofcancer cells,and were swallowed more than others by cancer cells.Conclusion CNPs have a promising application prospective in tumor-targeteddrug delivery.
