摘  要:以琴叶风吹楠2年生苗木叶片为试验材料,比较叶面积法和称重法,95%乙醇、80%丙酮、丙酮-乙醇(2∶1)、试剂盒等4种提取液,和叶样研磨、不研磨处理对提取叶绿素含量的影响,试验结果:叶面积法得到的叶绿素a、叶绿素b和叶绿素总含量与SPAD值的相关系数远大于称重法;在直接浸提条件下,叶面积法4种提取液得到叶绿素总含量均值分别为3.9686、5.9621、6.0023、6.5956 mg/dm2,称重法得到叶绿素总含量分别为1.7353、2.4773、2.6695、2.7669 mg/g,提取效果排序为试剂盒>丙酮-乙醇(2∶1)>80%丙酮>95%乙醇;混合提取液的提取效果均显著高于单一乙醇提取液,而混合提取液之间无显著差异;4种提取液提取研磨处理叶样的叶绿素总含量均值分别为6.088、6.513、6.429、7.278 mg/dm2,不研磨处理提取的叶绿素总含量分别为2.698、5.969、5.627、5.751 mg/dm2,研磨法提取效果较好。试验表明,用叶面积法,叶样研磨处理,以丙酮-乙醇(2∶1)为提取液的方法是较好的琴叶风吹楠叶绿素提取方法。Take2-year-old Horsfieldia pandurifolia as experiment material,the comparative effect of different methods on extractedchlorophyll content was studied to select the optimal one from leaf-area method and weighing method by using4extractionsolvents which represent95%ethanol,80%acetone,acetone-ethanol(2∶1)and kit,respectively under grinding and nogrinding treatments.The results showed that the contents of chlorophyll a,chlorophyll b and total chlorophyll content and thecorrelation of SPAD value obtained by leaf-area method are more precise than that of by weighing method,and the former correlationcoefficient is much more bigger than the latter;In the condition of direct soaking,the4kinds of extracts’average totalchlorophyll content obtained by the leaf-area method are3.9686,5.9621,6.0023and6.5956mg/dm2.And the corresponding resultsobtained by the weighing method are1.7353,2.4773,2.6695and2.7669mg/g,respectively.The mean total chlorophyllcontent of the4kinds of extracts by grinding are6.088,6.429,6.513,7.278mg/dm2,the corresponding results extract bythe wayof the no-grinding are2.698,5.969,5.627,5.751mg/dm2,it shows that grinding method has the better extraction efficiency.Inconclusion,it is effective to adapt leaf-area method with acetone ethanol(2∶1)solvent under grinding treatment to obtain thechlorophyll content of Horsfieldia pandurifolia.
