摘  要:目的观察大鼠脂多糖导致的急性肺损伤模型中NF-k B及低氧诱导因子-1a(HIF-1a)的表达及两者之间的相关性。方法通过往气管注入脂多糖建立急性肺损伤模型,然后于建模后的3、9、24h分别比较各组大鼠肺湿/干重比(W/D)、动脉血氧分压(PaO_2)、肺组织病理切片和肺损伤病理评分;免疫印迹法(Western blot)检测肺组织匀浆HIF-1a浓度、酶联免疫吸附(ELISA)法测定大鼠NF-k B含量,并将NF-k B及HIF-1a做相关性分析。结果 (1)ALI组3、9、24h PaO_2均较对照组明显降低(P<0.01)。(2)肺W/D、肺损伤组织病理评分:ALI组3、9、24h均较对照组明显升高(P<0.01),且在9h最高。(3)ALI组3、9、24h血清NF-k B较对照组明显升高(P<0.01),且在9h最高。(4)ALI组3、9、24h大鼠肺组织匀浆HIF-1a蛋白较对照组明显升高(P<0.01),且与血清NF-k B有明显相关性(r=0.931)。结论在脂多糖导致的急性肺损伤中,HIF-1a蛋白表达增加,与NF-k B等炎症因子相互作用,促进炎症的发生发展。Objective To study the relationship between NF-kB and HIF-1a in the lung tissue of acute lung injury rats induced by intratracheally injecting lipopolysaccharide(LPS).Methods Injected LPS through trachea to establish the model of acute lung injury,and detected PaO2and Wet/Dry weight(W/D),examined pathological changes of the lung tissue and ALI score at3,9and24hours after modeling.Measured serum NF-kB content by ELISA,observed the expression of HIF-1a in homogenate of lung tissue by Western blot,and analyzed the correlation of NF-kB and HIF-1a.Results PaO2decreased in ALI group at3,9and24h compared with control group(P<0.01).Compared with control group,W/D and ALI score in ALI group were higher at3,9and24h(P<0.01).And the highest was at9hour.And the level of NF-kB was higher in ALI group at3,9and24h than in control group(P<0.01).Compared with control group,HIF-1a in homogenate of lung tissue was higher in ALI group at3,9and24h(P<0.01),and the expression of HIF-1a protein and the NFkB was correlated positively(r=0.931).Conclusion In acute lung injury induced by LPS,the expression of HIF-1a in lung tissue of ALI rats increases,and interacts with NF-kB in the inflammatory process,and promotes the occurrence and development of acute lung injury.
